For a one year period, we measured soil fluxes of nitrous oxide (N2O), carbon dioxide (CO2) and methane (CH4), related soil parameters (temperature, water-filled pore space, mineral nitrogen content, N mineralization potential) and litterfall in two highly fertilized (250 kg N ha\u22121 year\u22121) coffee cultivation: a monoculture (CM) and a culture shaded by the N2 fixing legume species Inga densiflora (CIn). Nitrogen fertilizer addition significantly influenced N2O emissions with 84% of the annual N2O emitted during the post fertilization periods, and temporarily increased soil respiration and decreased CH4 uptakes. The higher annual N2O emissions from the shaded plantation (5.8 \u00b1 0.3 kg N ha\u22121 year\u22121) when compared to that from the monoculture (4.3 \u00b1 0.1 kg N ha\u22121 year\u22121) was related to the higher N input through litterfall (246 \u00b1 16 kg N ha\u22121 year\u22121) and higher potential soil N mineralization rate (3.7 \u00b1 0.2 mg N kg\u22121 d.w. d\u22121) in the shaded cultivation when compared to the monoculture (153 \u00b1 6.8 kg N ha\u22121 year\u22121 and 2.2 \u00b1 0.2 mg N kg\u22121 d.w. d\u22121). This confirms that the presence of N2 fixing shade trees can increase N2O emissions. This work reviewed seven important European LCA studies in a sensitivity analysis approach in order to get a better understanding of the roots of such a debate for three major biofuels in European production: rape methyl ester and ethanol from wheat and sugar beet. Global trends and variability of energy and GHG balances were depicted and completed with a sensitivity analysis carried out for each methodological and data parameter, which allowed making recommendations on the carrying out of LCA in a policy-making or a biofuels comparison context. Methodological choices, and especially allocation rule, appeared as key elements for results variation with influences on balances up to 149%; system expansion approach was identified as the most relevant rule since it integrates the market potential and the environmental interest of by-products promotion, which was pointed out as a crucial point for biofuels sustainability. The influence of local specificity for cultivation data was evaluated up to 167%, which puts too large geographical coverage in question. Modelling uncertainties due to N2O emissions from soils showed influences from 17 to 46%, which represents a crucial challenge for research and for LCA results accuracy. Approximations evaluation pointed out the need to integrate agricultural machinery into the assessment. Here, we report the genome sequencing of 39 accessions that were used to establish a white lupin pangenome. We defined 32\uffe2\uff80\uff89068 core genes that are present in all individuals and 14\uffe2\uff80\uff89822 that are absent in some and may represent a gene pool for breeding for improved productivity, grain quality, and stress adaptation. We used this new pangenome resource to identify candidate genes for alkaloid synthesis, a key grain quality trait. The white lupin pangenome provides a novel genetic resource to better understand how domestication has shaped the genomic variability within this crop. A. taxiformis contains several anti-methanogenic compounds including bromoform that inhibits directly methanogenesis. The positive and adverse effects of A. taxiformis on the rumen microbiota are dose-dependent and operate in a dynamic fashion. It is therefore key to characterize the dynamic response of the rumen microbial fermentation for identifying optimal conditions on the use of A. taxiformis as a dietary supplement for methane mitigation. Accordingly, the objective of this work was to model the effect of A. taxiformis supplementation on the rumen microbial fermentation under in vitro conditions. We adapted a published mathematical model of rumen microbial fermentation to account for A. taxiformis supplementation. We modelled the impact of A. taxiformis on the fermentation and methane production by two mechanisms, namely (i) direct inhibition of the growth rate of methanogens by bromoform and (ii) hydrogen control on sugars utilization and on the flux allocation towards volatile fatty acids production. We calibrated our model using a multi-experiment estimation approach that integrated experimental data with six macroalgae supplementation levels from a published in vitro study assessing the dose-response impact of A. taxiformis on rumen fermentation.</p>Results<p>our model captured satisfactorily the effect of A. taxiformis on the dynamic profile of rumen microbial fermentation for the six supplementation levels of A. taxiformis with an average determination coefficient of 0.88 and an average coefficient of variation of the root mean squared error of 15.2% for acetate, butyrate, propionate, ammonia and methane.</p>Conclusions<p>our results indicated the potential of our model as prediction tool for assessing the impact of additives such as seaweeds on the rumen microbial fermentation and methane production in vitro. Additional dynamic data on hydrogen and bromoform are required to validate our model structure and look for model structure improvements. In this study, we investigated the effect of vibro-ball milling (VBM) for 5 and 30 min at a frequency of 20 s\uffe2\uff88\uff921 on the physicochemical composition and enzymatic hydrolysis (30 U g\uffe2\uff88\uff921 total solids (TS) of cellulase and endo-1,4-xylanase from Trichoderma longibrachiatum) of dry and wet solid separated digestates from an agricultural biogas plant. We found that VBM of dry solid digestate improved the physical parameters as both the particle size and the crystallinity index (from 27% to 75%) were reduced. By contrast, VBM of wet solid digestate had a minimal effect on the physicochemical parameters. The best results in terms of cellulose and hemicelluloses hydrolysis were noted for 30 min of VBM of dry solid digestate, with hydrolysis yields of 64% and 85% for hemicelluloses and cellulose, respectively. At the condition of 30 min of VBM, bioethanol and methane production on the dry solid separated digestate was investigated. Bioethanol fermentation by simultaneous saccharification and fermentation resulted in an ethanol yield of 98 geth kg\uffe2\uff88\uff921 TS (corresponding to 90% of the theoretical value) versus 19 geth kg\uffe2\uff88\uff921 TS for raw solid digestate. In the present study, we tested the ability of Clostridium saccharoperbutylacetonicum N1-4 (DSMZ 14923) to produce acetate and D-3-hydroxybutyrate from inorganic carbon present in a CO2:N2 gas mix. At the same time, we tested the ability of a Shewanella oneidensis MR1 and Pseudomonas aeruginosa PA1430/CO1 consortium to provide reducing power to sustain carbon assimilation at the cathode. We tested the performance of three different systems with the same layouts, inocula, and media, but with the application of 1.5 V external voltage, of a 1000 \u2126 external load, and without any connection between the electrodes or external devices (open circuit voltage, OCV). We compared both CO2 assimilation rate and production of metabolites (formate, acetate 3-D-hydroxybutyrate) in our BESs with the values obtained in non-electrogenic control cultures and estimated the energy used by our BESs to assimilate 1 mol of CO2. Our results showed that C. saccharoperbutylacetonicum NT-1 achieved the maximum CO2 assimilation (95.5%) when the microbial fuel cells (MFCs) were connected to the 1000 \u2126 external resistor, with the Shewanella/Pseudomonas consortium as the only source of electrons. Furthermore, we detected a shift in the metabolism of C. saccharoperbutylacetonicum NT-1 because of its prolonged activity in BESs. In the present study, we tested the ability of Clostridium saccharoperbutylacetonicum N1-4 (DSMZ 14923) to produce acetate and D-3-hydroxybutyrate from inorganic carbon present in a CO2:N2 gas mix. At the same time, we tested the ability of a Shewanella oneidensis MR1 and Pseudomonas aeruginosa PA1430/CO1 consortium to provide reducing power to sustain carbon assimilation at the cathode. We tested the performance of three different systems with the same layouts, inocula, and media, but with the application of 1.5 V external voltage, of a 1000 \u2126 external load, and without any connection between the electrodes or external devices (open circuit voltage, OCV). We compared both CO2 assimilation rate and production of metabolites (formate, acetate 3-D-hydroxybutyrate) in our BESs with the values obtained in non-electrogenic control cultures and estimated the energy used by our BESs to assimilate 1 mol of CO2. Our results showed that C. saccharoperbutylacetonicum NT-1 achieved the maximum CO2 assimilation (95.5%) when the microbial fuel cells (MFCs) were connected to the 1000 \u2126 external resistor, with the Shewanella/Pseudomonas consortium as the only source of electrons. Furthermore, we detected a shift in the metabolism of C. saccharoperbutylacetonicum NT-1 because of its prolonged activity in BESs.
