The newly produced PPI and PF fractions were processed into pasta-like sheets with varying protein to fiber ratios (100/0, 90/10, 80/20, 70/30 and 50/50, respectively) using high temperature compression molding. We studied protein polymerization, molecular structure and protein-fiber interactions, as well as mechanical performance and cooking characteristics of processed PPI-PF blends. Bi-modal particle size distribution and chemical composition of the PPI and PF fractions influenced significantly the physicochemical properties of the pasta-like sheets. Polymerization was most pronounced for the 100 PPI, 90/10 and 80/20 PPI-PF samples as studied by SE-HPLC, and polymerization decreased with addition of the PF fraction. The mechanical properties, as strength and extensibility, were likewise the highest for the 100 PPI and 90/10 PPI-PF blends, while the E-modulus was similar for all the studied blends (around 38\u202fMPa). The extensibility decreased with the increasing amount of PF in the blend. The highest amounts of \u03b2-sheets were found in the pasta-like sheets with high amounts of PPI (100, 90 and 80%), by FT-IR. An increase in PF fraction in the blend, resulted into the high amounts of unordered structures as observed by FT-IR, as well as in an increase in the molecular scattering distances observed by SAXS. The water uptake increased and cooking loss decreased with increased proportions of the PF fraction, and the consistency of 10\u202fmin cooked pasta-like sheets were alike al dente texture. The new knowledge obtained in this study on the use of extraction-fractionation method to produce novel PPI and PF fractions for developing innovative high nutritious food can be of a great importance. The obtained knowledge on the pea protein and fiber processing behaviour could greatly contribute to a better control of functional properties of various temperature-processed products from yellow pea.", "keywords": ["Dietary Fiber", "2. The trial is a long\uffe2\uff80\uff90term field experiment which divides conventional and organic agriculture into component pest\uffe2\uff80\uff90control and soil treatment practices. We hypothesised that management practices would affect soil ecology and used community level physiological profiles, microbial and nematode counts, and denaturing gradient gel electrophoresis (DGGE) to characterise soil microbial communities in plots used for onion (Allium cepa L.) cultivation.</p>RESULTS<p>Microbial activity and culturable bacterial counts were significantly higher under fully organic management. Culturable fungi, actinomycete and nematode counts showed a consistent trend towards higher numbers under fully organic management but these data were not statistically significant. No differences were found in the fungal/bacterial ratio. DGGE banding patterns and sequencing of excised bands showed clear differences between treatments. Putative onion fungal pathogens were predominantly sequenced under conventional soil treatment practices whilst putative soil suppressive bacterial species were predominantly sequenced from the organic pest\uffe2\uff80\uff90control treatment plots.</p>CONCLUSION<p>Organic management increased microbial activity and diversity. Excess deposition of reactive nitrogen is a well-known agent of stress for lichens, but which symbiont is most affected and how, remains a mystery. Using proteomics can expand our understanding of stress effects on lichens. We investigated the effects of different doses and forms of reactive nitrogen, with and without supplementary phosphorus and potassium, on the proteome of the lichen Cladonia portentosa growing in a 'real-world' simulation of nitrogen deposition. The photobiont mainly responded overexpressing proteins involved in energy production. Because soil microorganisms depend on plant litter as a source of energy for metabolism, changes in the amount or the biochemistry of plant litter produced under elevated CO(2) and O(3) could alter microbial community function and composition. Previously, we have observed that elevated CO(2) increased the microbial metabolism of cellulose and chitin, whereas elevated O(3) dampened this response. We hypothesized that this change in metabolism under CO(2) and O(3) enrichment would be accompanied by a concomitant change in fungal community composition. We tested our hypothesis at the free-air CO(2) and O(3) enrichment (FACE) experiment at Rhinelander, Wisconsin, in which Populus tremuloides, Betula papyrifera, and Acer saccharum were grown under factorial CO(2) and O(3) treatments. We employed extracellular enzyme analysis to assay microbial metabolism, phospholipid fatty acid (PLFA) analysis to determine changes in microbial community composition, and polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) to analyze the fungal community composition. The activities of 1,4-beta-glucosidase (+37%) and 1,4,-beta-N-acetylglucosaminidase (+84%) were significantly increased under elevated CO(2), whereas 1,4-beta-glucosidase activity (-25%) was significantly suppressed by elevated O(3). There was no significant main effect of elevated CO(2) or O(3) on fungal relative abundance, as measured by PLFA. We identified 39 fungal taxonomic units from soil using DGGE, and found that O(3) enrichment significantly altered fungal community composition. We conclude that fungal metabolism is altered under elevated CO(2) and O(3), and that there was a concomitant change in fungal community composition under elevated O(3). Soils from thirteen accredited organic farms and conventionally managed neighboring farms were analyzed using a polyphasic approach combining traditional soil analysis, culture-dependent and independent microbiological analyses, a nematode community analysis and an enquiry about different management practices among the farmers. Organic management, known primarily for the abstinence of artificial fertilizers and pesticides, resulted in significantly lower levels of both nitrate and total soluble nitrogen in the soil, higher numbers of bacteria of different trophic groups, as well as larger species richness in both bacteria and nematode communities and more resilience to a drying\u2013rewetting disturbance in the soil. The organic farmers plough their fields less deeply and tend to apply more organic carbon to their fields, but this did not result in a significantly higher organic carbon content in their soils. The levels of ammonium, organic nitrogen, phosphate and total phosphorus did not differ, significantly between the soils under different management. Fifty percent of the conventional Dutch farmers also used organic fertilizers and the numbers of farmers using a green crop fertilizer did not differ between the two management types. Soil type \u2013 clayey or sandy soil \u2013 in general had a much stronger effect on the soil characteristics than management type. The soil type influenced pH, nitrate, ammonium, phosphate and organic carbon levels as well as numbers of oligotrophic bacteria and of different groups of nematodes, and different diversity indices. Therefore, in the present study, a wide-range screening of the fungal endophyte communities was conducted to determine the fungal distribution and diversity on S. caprea roots on a metal polluted site. Fungal communities were screened using amplification with the 5.8S-ITS2-28S part of the rDNA operon, with the resulting amplicons analysed by temporal temperature gradient gel electrophoresis (TTGE) and sequencing. This technique is reproducible and shows good coverage of ascomycete and basidiomycete taxa, as 68% and 32% of all of the sequences, respectively. No clear shift in fungal ITS-TTGE profiles from S. caprea roots was seen along the secondary succession stages. Ascomycetes dominated the more polluted plots, while there was a greater diversity of basidiomycetes in the less polluted and control plots, suggesting greater tolerance of ascomycetes in comparison with basidiomycete fungi. The high diversity of DSEs was confirmed at the highly metal-enriched locations, with species belonging to the genera Phialophora, Phialocephala and Leptodontidium. Consequently, we carried out a 4-month microcosm experiment to measure the impact of different sources of environmental degradation (biodiversity loss, nitrogen deposition and climate change) on soil health in a non-polluted (non-degraded) and a heavily metal-polluted (degraded) soil, and to compare their responses. To this aim, we determined a variety of soil microbial properties with potential as bioindicators of soil health: basal respiration; \u03b2-glucosaminidase and protease activities; abundance (Q-PCR) of bacterial, fungal and chitinase genes; richness (PCR-DGGE) of fungal and chitinase genes. Non-polluted and metal-polluted soils showed different response microbial dynamics when subjected to sources of environmental degradation. The non-polluted soil appeared resilient to 'biodiversity loss' and 'climate change' treatments. The metal-polluted soil was probably already too severely affected by the presence of high levels of toxic metals to respond to other sources of stress. Recent developments in molecular biology based techniques have led to rapid and reliable tools to characterize microbial community structures. We determined the effects of conventional and no-tillage in cropping systems with and without cover crops on bacterial community structure, total organic carbon (TOC) and soil aggregation. Tillage and rotation did not affect TOC from bulk soil. However, TOC was greater in the largest aggregate size class (7.98-19 mm), and had greater mean-weight diameter under no-tillage than under conventional tillage in the 0-5 cm soil layer. Barley (Hordeum vulgare L. cv. Optic) and maize (Zea mays L. cv. Freya) root exudates were collected using an aerated hydroponic method and compared with chia (Salvia hispanica L.) seed exudate, a commonly used root exudate analogue. Sandy loam soil was passed through a 500\uffe2\uff80\uff90\uffce\uffbcm mesh and treated with each exudate at a concentration of 4.6 mg exudate g\uffe2\uff88\uff921 dry soil. Two sets of soil samples were prepared. One set of treated soil samples was maintained at 4\uffc2\uffb0C to suppress microbial processes. To characterize the effect of decomposition, the second set of samples was incubated at 16\uffc2\uffb0C for 2 weeks at \uffe2\uff88\uff9230 kPa matric potential. Gas chromatography\uffe2\uff80\uff93mass spectrometry (GC\uffe2\uff80\uff93MS) analysis of the exudates showed that barley had the largest organic acid content and chia the largest content of sugars (polysaccharide\uffe2\uff80\uff90derived or free), and maize was in between barley and chia. Yield stress of amended soil samples was measured by an oscillatory strain sweep test with a cone plate rheometer. When microbial decomposition was suppressed at 4\uffc2\uffb0C, yield stress increased 20\uffe2\uff80\uff90fold for chia seed exudate and twofold for maize root exudate compared with the control, whereas for barley root exudate decreased to half. The yield stress after 2 weeks of incubation compared with soil with suppressed microbial decomposition increased by 85% for barley root exudate, but for chia and maize it decreased by 87 and 54%, respectively. Barley root exudation might therefore disperse soil and this could facilitate nutrient release. We observed that the main differences in the bacterial communities were not related to land-use type, but rather to soil factors. An exception was the bacterial community of pine forest soils (PFS), which was clearly different from all other sites. PFS had lowest bacterial abundance, lowest numbers of operational taxonomic units (OTUs), lowest soil pH, and highest C : N ratios. C : N ratio strongly influenced bacterial community structure and was the main factor separating PFS from other fields. For the sites other than PFS, phosphate was the most important factor explaining the differences in bacterial communities across fields. Firmicutes were the most dominant group in almost all fields, except in PFS and deciduous forest soils (DFS). In PFS, Alphaproteobacteria was most represented, while in DFS, Firmicutes and Gammaproteobacteria were both highly represented. The site has been intensively studied to measure the impact of stubble management and N-fertilizer application on greenhouse gas emissions (CO(2) and N(2)O), N-cycling, pathology, soil structure and yield. As all of these endpoints can be regulated by microbial processes, the microbiology of the system was examined. Soil samples were taken after a winter fallow period and the diversity of the bacterial and fungal communities was measured using PCR-denaturing gradient gel electrophoresis. Stubble and N shifted the structure of bacterial and fungal communities with the primary driver being stubble addition on the fungal community structure (P<0.05 for all effects). Changes in C, N (total and NO(3)), K and Na, were correlated (P<0.05) with variation in the microbial community structure. Quantitative PCR showed that nifH (nitrogen fixation) and napA (denitrification) gene abundance increased upon stubble retention, whereas amoA gene numbers were increased by N addition. Domestic herbivore grazing is known to influence soil bacterial community. However, the effects of grazing and its major driving factors on soil bacterial community remain unknown for different plant community compositions under increasing grazing intensity. Thus, to investigate soil bacterial community diversity under five plant community compositions (Grass; Leymus chinensis; Forb; L. chinensis & Forb; and Legume), we performed a four-year field experiment with different grazing intensity treatments (no grazing; light grazing, 4 sheep\u00b7ha-1; and heavy grazing, 6 sheep\u00b7ha-1) in a grassland in China. Total DNA was obtained from soil samples collected from the plots in August, and polymerase chain reaction (PCR) analysis and denaturing gradient gel electrophoresis (DGGE) fingerprinting were used to investigate soil bacterial community. The results showed that light grazing significantly increased indices of soil bacterial community diversity for the Forb and Legume groups but not the Grass and L. chinensis groups. Heavy grazing significantly reduced these soil bacterial diversity indices, except for the Pielou evenness index in the Legume group. Further analyses revealed that the soil N/P ratio, electrical conductivity (EC), total nitrogen (TN) and pH were the major environmental factors affecting the soil bacterial community. Our study suggests that the soil bacterial community diversity was influenced by grazing intensity and plant community composition in a meadow steppe. Barley (                       Hordeum vulgare                       L. cv. Optic) and maize (                       Zea mays                       L. cv. Freya) root exudates were collected using an aerated hydroponic method and compared with chia (                       Salvia hispanica                       L.) seed exudate, a commonly used root exudate analogue. Sandy loam soil was passed through a 500                       \uffe2\uff80\uff90                       \uffce\uffbcm mesh and treated with each exudate at a concentration of 4.6 mg exudate g                       \uffe2\uff88\uff921                       dry soil. Two sets of soil samples were prepared. One set of treated soil samples was maintained at 4\uffc2\uffb0C to suppress microbial processes. To characterize the effect of decomposition, the second set of samples was incubated at 16\uffc2\uffb0C for 2 weeks at \uffe2\uff88\uff9230 kPa matric potential. Yield stress of amended soil samples was measured by an oscillatory strain sweep test with a cone plate rheometer. When microbial decomposition was suppressed at 4\uffc2\uffb0C, yield stress increased 20\uffe2\uff80\uff90fold for chia seed exudate and twofold for maize root exudate compared with the control, whereas for barley root exudate decreased to half. The yield stress after 2 weeks of incubation compared with soil with suppressed microbial decomposition increased by 85% for barley root exudate, but for chia and maize it decreased by 87 and 54%, respectively. Barley root exudation might therefore disperse soil and this could facilitate nutrient release. The results showed that, for a given range, the addition of water glass consumed the Ca(OH)2 in samples and generated large amounts of calcium silicate hydrate (C-S-H) gels, forming a compact microstructure, which was beneficial for the rapid development of strength. The procedure can be performed using minigel electrophoresis systems and does not involve preparation of gradient-separating gels or the use of special cooling devices. The procedure accommodates both silver and Coomasie Blue staining. Thus the procedure is simple to perform and highly repeatable, providing high-resolution separation of MHC protein isoforms. The percent composition of the four adult MHCs in rat soleus, medial gastrocnemius, diaphragm, and levator ani muscles by use of this procedure and Coomasie Blue staining is similar to that previously reported.
