Our aim was to test this hypothesis for intercropped durum wheat and faba bean along a P-fertilizer gradient. A field experiment was conducted in a long-term P-fertilizer trial with three rates of P-fertilization (No, Low and High P). Microbial biomass was assessed by chloroform fumigation-extraction. Quantitative PCR was applied to evaluate the abundance of relevant microbial groups. Phosphorus availability and microbial biomass systematically increased in the rhizosphere compared to bulk soil. P-fertilization resulted in higher abundance of targeted bacterial phyla, whole bacterial and fungal communities, and depressed mycorrhizal colonization of durum wheat, but not faba bean. Microbial biomass carbon significantly increased in the rhizosphere only in P-fertilized treatments, pointing to P limitation of microbial communities. Intercropping yielded a significant effect on rhizosphere microbial properties only at High P. Microbial biomass P increased in the rhizosphere of intercropped faba bean only at No P level, and was thus the sole finding supporting the stress-gradient hypothesis. P-fertilization was the main driver of microbial communities in this field trial, and P-fertilizer application modulated the species-specific effect in the intercrop. Due to the extensive use and high resistance to degradative factors, these compounds pose a significant risk of environmental spreading, bioaccumulating also in living organisms. In this context, despite many researches have been performed to demonstrate 'legacy' PFAS harmfulness, only few data are still available about all the emerging fluorinated molecules, industrially introduced to replace the previous ones. For this reason, we proposed the medicinal leech Hirudo verbana as consolidated invertebrate model to assess the effects of four different PFAS (HFPO-DA, PFMoBa, PFOA and PFMOPrA) following freshwater dispersion. Morphological, immunohistochemical and molecular analyses demonstrate that, despite all the compounds basically induce an acute inflammatory and oxidative stress response, a different cellular and molecular response has been observed. Whereas for PFOA and PFMOPrA an increase in the tested concentration leads to a corresponding rise in the immune response, HFPO-DA and PFMoBa trigger an entirely opposite effect. Indeed, the significant recruitment of both granulocytes and macrophage like cells, typically involved in the removal of non-self, is inhibited with increasing concentrations of these compounds. A second bacteria and bromide pulse was then poured on the column surfaces and leaching was extended six more pore volumes. Attachment by either bacteria decreased markedly in water-repellent sand, so straining was the main retention mechanism. This pilot study investigates microplastic pollution from a mixed plastics recycling facility in the UK to advance current quantitative understanding of microplastic (MP) pollution release from a plastic recycling facility to receiving waters. Raw recycling wash water were estimate to contain microplastic counts between 5.97\u00d7 106 \u2013 1.12 \u00d7 108 MP m\u22123 (following fluorescence microscopy analysis) . The microplastic pollution mitigation (filtration installed) was found to remove the majority of microplastics >5\u03bcm, with high removal efficiencies for microplastics >40\u03bcm. Microplastics <5\u03bcm were generally not removed by the filtration and subsequently discharged, with 59-1184 tonnes potentially discharged annually. It is recommended that additional filtration to remove the smaller microplastics prior to wash discharge is incorporated in the wash water management. Evidence of microplastic wash water pollution suggest it may be important to integrate microplastics into water quality regulations. This study presents the development and validation of a new, highly sensitive method for the quantification of 32 PFAS in human serum using online solid-phase extraction (SPE) coupled with ultra-high performance liquid chromatography-high resolution mass spectrometry (UHPLC-HRMS). Legacy and emerging PFAS were targeted. Main steps of sample pretreatment include protein precipitation (PP), pellet rinsing, centrifugation, preconcentration through solvent evaporation, and online SPE using a weak anion-exchange polymeric sorbent. The PP and pellet-rinsing procedures were optimized through a comprehensive exploration of solvent combinations. Following this, a pretreatment that offers the best compromise for the targeted PFAS was identified using principal component analysis. The method demonstrated excellent linearity (R\u00b2 = 0.977-0.997) with limits of quantification ranging from 8.9 to 27\u00a0ng/L, 5 to 15 times lower than previous methods. Precision (intraday 2.6-14.0\u00a0% and interday 1.3-11.0\u00a0% relative standard deviation) and accuracy (recoveries 72.7-106\u00a0%) were robust. The method was validated in accordance with ISO/IEC 17025 and successfully applied to five human serum samples, confirming its suitability for high-throughput profiling of PFAS in biomonitoring studies. This method is the first to use online SPE for the simultaneous determination of a broad range of PFAS, including ether congeners such as perfluoro(2-ethoxyethane) sulfonic acid and Nafion byproduct 2. In-situ soil flushing with solvents has been considered as a remediation technique for PFAS-contaminated soils. The use of non-Newtonian fluids, displaying variable viscosity depending on the applied shear rate, can offer certain advantages in improving the efficiency of the process, particularly in heterogeneous porous media. In this work, the efficacy of ethanol/xanthan mixture (XE) in the recovery of a mixture of perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorohexane sulfonate (PFHxS), and perfluorobutane sulfonate (PFBS) from soil has been tested at lab-scale. XE's non-Newtonian behavior was examined through rheological measurements, confirming that ethanol did not affect xanthan gum's (XG) shear-thinning behavior. The recovery of PFAS in batch-desorption exceeded 95\u00a0% in ethanol, and 99\u00a0% in XE, except for PFBS which reached 94\u00a0%. 1D-column experiments revealed overshoots in PFAS breakthrough curves during ethanol and XE injection, due to over-solubilization. XE, (XG 0.05\u00a0% w/w) could recover 99\u00a0% PFOA, 98\u00a0% PFBS, 97\u00a0% PFHxS, and 92\u00a0% PFOS. Experiments were carried out using a submerged polyethersulfone ultrafiltration membrane with molecular weight cutoff of 100 kDa in combination with air stripping and addition to acid and base traps for recovery of volatiles. Results showed, that the best operation mode of ultrafiltration is with backwash cycles of the permeate, maintaining a flux lower than the critical flux of 6 L/h\u00b7m<sup>2</sup>. The setup led to fast stripping of ammonia, which was successfully recovered by 88% in the acid trap. This application can be considered an adequate first stage treatment of the HTL wastewater. This study presents the development and validation of a new, highly sensitive method for the quantification of 32 PFAS in human serum using online solid-phase extraction (SPE) coupled with ultra-high performance liquid chromatography-high resolution mass spectrometry (UHPLC-HRMS). Legacy and emerging PFAS were targeted. Main steps of sample pretreatment include protein precipitation (PP), pellet rinsing, centrifugation, preconcentration through solvent evaporation, and online SPE using a weak anion-exchange polymeric sorbent. The PP and pellet-rinsing procedures were optimized through a comprehensive exploration of solvent combinations. Following this, a pretreatment that offers the best compromise for the targeted PFAS was identified using principal component analysis. The method demonstrated excellent linearity (R\u00b2 = 0.977-0.997) with limits of quantification ranging from 8.9 to 27\u00a0ng/L, 5 to 15 times lower than previous methods. Precision (intraday 2.6-14.0\u00a0% and interday 1.3-11.0\u00a0% relative standard deviation) and accuracy (recoveries 72.7-106\u00a0%) were robust. The method was validated in accordance with ISO/IEC 17025 and successfully applied to five human serum samples, confirming its suitability for high-throughput profiling of PFAS in biomonitoring studies. In this context, despite many researches have been performed to demonstrate 'legacy' PFAS harmfulness, only few data are still available about all the emerging fluorinated molecules, industrially introduced to replace the previous ones. For this reason, we proposed the medicinal leech Hirudo verbana as consolidated invertebrate model to assess the effects of four different PFAS (HFPO-DA, PFMoBa, PFOA and PFMOPrA) following freshwater dispersion. Morphological, immunohistochemical and molecular analyses demonstrate that, despite all the compounds basically induce an acute inflammatory and oxidative stress response, a different cellular and molecular response has been observed. Raman spectroscopy was most sensitive to soil organic matter residues, requiring more sophisticated sample pretreatment. Fluorescence staining with subsequent Fluorescence microscopy detection effectively recovered most small-sized LDPE-MiP but missed 56\u201393 % of small PBAT/ PLA particles. For the latter, reliable quantification was achieved only using Soxhlet extraction combined with 1H NMR spectroscopic quantification. Pyrolysis-GC-MS showed intermediate results, displaying low sensitivity to plastic type and lower recoveries as soil clay content increased. We conclude that different methods have different sensitivities for different MiP materials in different soils, i.e. comparisons of MiP loads and threshold settings for MiP loads across methodologies require careful consideration. We used a controlled experimental setup to explore hydrophilic Escherichia coli (E. coli) and hydrophobic Rhodococcus erythropolis (PTCC1767) (R. erythropolis) transport through dry (-\u00a015,000\u00a0cm water potential) and water saturated (0\u00a0cm water potential) wettable and water-repellent sand columns. A pulse of bacteria (1\u00a0\u00d7\u00a0108 CFU mL-1) and bromide (10\u00a0mmol\u00a0L-1) moved through the columns under saturated flow (0\u00a0cm) for four pore volumes. A second bacteria and bromide pulse was then poured on the column surfaces and leaching was extended six more pore volumes. In dry wettable sand attachment dominated E. coli retention, whereas R. erythropolis was dominated by straining. Once wetted, the dominant retention mechanisms flipped between these bacteria. Attachment by either bacteria decreased markedly in water-repellent sand, so straining was the main retention mechanism. In-situ soil flushing with solvents has been considered as a remediation technique for PFAS-contaminated soils. The use of non-Newtonian fluids, displaying variable viscosity depending on the applied shear rate, can offer certain advantages in improving the efficiency of the process, particularly in heterogeneous porous media. In this work, the efficacy of ethanol/xanthan mixture (XE) in the recovery of a mixture of perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorohexane sulfonate (PFHxS), and perfluorobutane sulfonate (PFBS) from soil has been tested at lab-scale. XE's non-Newtonian behavior was examined through rheological measurements, confirming that ethanol did not affect xanthan gum's (XG) shear-thinning behavior. The recovery of PFAS in batch-desorption exceeded 95\u00a0% in ethanol, and 99\u00a0% in XE, except for PFBS which reached 94\u00a0%. 1D-column experiments revealed overshoots in PFAS breakthrough curves during ethanol and XE injection, due to over-solubilization. XE, (XG 0.05\u00a0% w/w) could recover 99\u00a0% PFOA, 98\u00a0% PFBS, 97\u00a0% PFHxS, and 92\u00a0% PFOS.
