The objective of this acidification is to increase the amount of phosphorus available in the digestate liquid fraction and, subsequently, recover this element by chemical precipitation in the form of struvite or calcium phosphate. Two digestate samples (one fresh and one old) were studied and treated by adding various amounts of sulphuric acid to the different digestate fractions (raw digestate, solid fraction and liquid fraction). For the raw digestate, phosphorus releases higher than 95% were obtained for pH 4.0. In the last part of the experiment, the influence of acid pre-treatment on the reaction yield of phosphorus precipitation, in the form of struvite or calcium phosphate, was determined. This study explores the application of portable membrane inlet mass spectrometer (MIMS) for noninvasive hive analysis, demonstrating its ability to detect various compounds indicative of hive conditions and environmental stressors. In addition to the expected compounds found in beehives, such as \u03b1-bergamotene, hexadecanoic acid, heptadecane, hexadecanamide,\u00a0\u03b1-bisabolol-, 9-octadecenamide, (Z)\u2009\u2212\u2009, and benzaldehyde, unexpected compounds, pollutants, like indane (polycyclic aromatic hydrocarbon) and carbofuran (pesticide), were also detected. The MIMS detection method provides rapid, accurate, and real-time results, making it suitable for preventive measures against bee diseases and integral to environmental biomonitoring. Soil samples were collected and analyzed from Compost (cattle manure, 2,000 kg/10a), 1/2 NPK+compost (14-20-14+2,000 kg/10a), NPK+compost (28-40-28+2,000 kg/10a), NPK (28-40-28 kg/10a), 3 NPK (84-120-84 kg/10a), and Control (no fertilization) plot which have been managed in the same manners with compost and different amount of chemical fertilization. The range of pyrosequencing reads and OTUs were 4,687-7,330 and 1,790-3,695, respectively. Species richness estimates such as Ace, Chao1, and Shannon index were higher in 1/2 NPK+compost than other treatments, which were 15,202, 9,112, 7.7, respectively. Dominant bacterial groups at level of phylum were Proteobacteria, Acidobacteria, and Actinobacteria. Those were occupied at 70.9% in 1/2 NPK+compost. Dominant bacterial groups at level of genus were Pseudolabrys, Bradyrhizobium, and Acidobacteria. Those were distributed at 14.4% of a total of bacteria in Compost. Soil pH displayed significantly closely related to bacterial species richness estimates such as Ace, Chao1 (p<0.05) and Shannon index (p<0.01). However, it showed the negative correlation with exchangeable aluminum contents (p<0.05). We selected three representative sites (3\u00a0ha each) with low, moderate and high grazing disturbance located far, mid-distance and near the watering point, respectively, in rangelands submitted to sheep grazing for more than 100 years. We assessed the canopy structure and identified the four most frequent plant patch types at each site. We selected four replications of each patch type and extracted a soil sample (0-30\u00a0cm depth) underneath the canopy and in the middle of the nearest inter-patch bare soil area in winter and summer. We assessed the root and soil dry mass and the respective organic C concentration in each sample and then we estimated the total belowground organic C storage at each site. Total plant and perennial grass cover were lower with high than low grazing disturbance while the reverse occurred with dwarf shrub cover. High grazing disturbance led to the increase in total root biomass in the whole soil profile of patch areas and in the upper soil of inter-patch areas. SOC was higher in patch than in inter-patch areas at all sites but at both areas was reduced with high grazing disturbance. This was probably the result of the low total plant cover and the low and recalcitrant contribution of above and below-ground plant litter to soils at sites with high grazing disturbance. Accordingly, these changes did not result in variations in the total belowground organic C storage. We concluded that high grazing disturbance did not affect the total belowground organic C storage but led to changes in the spatial patterning of this organic C storage (i.e shifting from soil to roots).", "keywords": ["0106 biological sciences", "2. Due to the extensive use and high resistance to degradative factors, these compounds pose a significant risk of environmental spreading, bioaccumulating also in living organisms. In this context, despite many researches have been performed to demonstrate 'legacy' PFAS harmfulness, only few data are still available about all the emerging fluorinated molecules, industrially introduced to replace the previous ones. For this reason, we proposed the medicinal leech Hirudo verbana as consolidated invertebrate model to assess the effects of four different PFAS (HFPO-DA, PFMoBa, PFOA and PFMOPrA) following freshwater dispersion. Morphological, immunohistochemical and molecular analyses demonstrate that, despite all the compounds basically induce an acute inflammatory and oxidative stress response, a different cellular and molecular response has been observed. Whereas for PFOA and PFMOPrA an increase in the tested concentration leads to a corresponding rise in the immune response, HFPO-DA and PFMoBa trigger an entirely opposite effect. Indeed, the significant recruitment of both granulocytes and macrophage like cells, typically involved in the removal of non-self, is inhibited with increasing concentrations of these compounds. Mono-digestion reduced most impacts, but represented a limited source for bio-energy. The easy-to-use, on-site, on-demand, and rapid monitoring of mycotoxins in food/feed is highly desired. In this work, we report on an advanced mycotoxin biosensor based on an array of graphene field-effect transistors integrated on a single silicon chip. A specifically designed aptamer against Ochratoxin A (OTA) was used as a recognition element, where it was covalently attached to graphene surface via pyrenebutanoic acid, succinimidyl ester (PBASE) chemistry. Namely, an electric field stimulation was used to promote more efficient \u03c0-\u03c0 stacking of PBASE to graphene. The specific G-rich aptamer strand suggest its \u03c0-\u03c0 stacking on graphene in free-standing regime and reconfiguration in G-quadruplex during binding an OTA molecule. This realistic behavior of the aptamer is sensitive to the ionic strength of the analyte solution, demonstrating a 10-fold increase in sensitivity at low ionic strengths. The graphene-aptamer sensors reported here demonstrate fast assay with the lowest detection limit of 1.4 pM for OTA within a response time as low as 10 s, which is more than 30 times faster compared to any other reported aptamer-based methods for mycotoxin detection. The treatments consisted of control, no input; MSWC, at 60 kgNha(-1); well decomposed cow manure (DCM), at 60 kgNha(-1); MSWC (30 kgNha(-1)) +Urea (30 kgNha(-1)); DCM (30 kgNha(-1)) +U (30 kgNha(-1)) and Fertilizer, (at 60:30:30 NPK kgha(-1) through urea, single superphosphate and muriate of potash respectively). Soil microbial biomass-C (MBC), MBC as percentage of organic-C (ratio index value, RIV), urease and acid phosphatase activities were higher in DCM than MSWC-treated soils, due to higher amount of biogenic organic materials like water soluble organic carbon, carbohydrate and mineralizable nitrogen in the former. The studied parameters were higher when urea was integrated with DCM or MSWC, compared to their single applications. Soil MBC, urease and acid phosphatase activities periodically declined up to 60 day after transplanting (DAT) and then increased after crop harvest. Soils from military range and training areas (RTAs) are contaminated principally by energetic materials (EM) and metals. Their chemical characteristics are relatively well known and toxicity assessment of soils from RTAs are in some cases available. However, bioavailability on these sites needs to be comprehensively characterized. A holistic approach to bioavailability, incorporating both chemical and earthworm toxicological indicators, was applied to soils from an anti-tank firing range at a Canadian Forces Base. Results showed that HMX and the metals Zn, Pb, Bi and Cd, though not consistently the prevailing toxicants, were the most accessible to earthworms. Some metals (notably Cu, Zn, Cr and Bi) were also accumulated in earthworm tissue but those were not necessarily expected given their bioaccessibility (i.e., the chemical availability of contaminants in the environment for the organisms) at the beginning of the exposure. The tested soils impaired earthworm reproduction and reduced adult growth. Measurement of selected sublethal parameters indicated that lysosomal integrity (determined as the neutral red retention time--NRRT) was decreased, while elevated superoxide dismutase (SOD) activity suggested that earthworms experienced oxidative stress. The correspondence between the NRRT and metal contamination pattern suggested that metals may be the main cause of lysosomal disruption in EM-contaminated soils. Rate of Se volatilization by wheat and rapeseed crops increased by 1.8-4.0 times; the greatest increase was observed with press mud followed by poultry manure, arhar (Cajanus cajan) leaves and farmyard manure. After 134d of incubation of 500g soil amended with 2% of plant tissues, the maximum amount of Se was volatilized with cowpea leaves (385ng) followed by wheat grains, leaves of maize, sugarcane (Saccharum officcinarum), arhar, poplar (Populus deltoides) and the control (91ng). In order to evaluate the effect of different sources (BS) and particle sizes (BF) of biochar on C loss and/or stability in soils, an incubation study on C mineralization of different biochar sources and particle sizes was established using two soils (ST): Norfolk soil (fine loamy, kaolinitic, thermic, typic Kandiudults) and Coxville soil (fine loamy kaolinitic, thermic, Paleaquults). In separate incubation vessels, these soils were amended with one of two manure-based biochars (poultry litters, PL; swine solids, SS) or one of two lignocellulosic-based biochars (switchgrass, SG; pine chips, PC) which were processed into two particle sizes (dust, <0.42 mm; pellet, >2 mm). The amount of CO2 evolved varied significantly between soils (p\u22640.0001); particle sizes (p\u22640.0001) and the interactions of biochar source (p\u22640.001) and forms of biochars (p\u22640.0001) with soil types. Averaged across soils and sources of biochar, CO2-C evolved from dust-sized biochar (281 mg kg(-1)) was significantly higher than pellet-sized biochar (226 mg kg(-1)). Among environmental pollutants, polystyrene (PS) stands out as a primary constituent of environmental plastic waste, given its widespread use and high production rates owing to its durability and user-friendly properties. The detection of polystyrene microparticles (PS-MPs) in various living organisms has been well-documented, posing a serious threat due to their potential passage into the human ecosystem. In this manuscript, we aimed to study the toxicological effects of low concentrations of pristine and photoaged PS-MPs in a murine macrophage cell line. To this purpose, PS-MPs were photoaged by indoor exposure to visible light to simulate environmental weathering due to solar irradiation (PS-MPs3h). Physical characterization revealed that the irradiation treatment results in particle degradation and the possible release of nanoparticles. Monocultures of the RAW264.7\u00a0cell line were then exposed to PS-MPs and PS-MPs3h at concentrations comparable to experimental measurements from biological samples, to assess cytotoxicity, intracellular oxidative stress, primary genotoxicity, and inflammatory effects. Significant toxicity-related outcomes were observed in cells treated with both pristine PS-MPs and PS-MPs3h even at low concentrations (0,10\u00a0\u03bcg/ml and 1\u00a0\u03bcg/ml). This study used mussel shell, cow bone, and biochar to reduce lead (Pb) toxicity in the highly contaminated military shooting range soil in Korea. Water-soluble and 1-M ammonium nitrate extractions, and a modified physiologically based extraction test (PBET) were performed to determine Pb bioavailability and bioaccessibility in the soil, respectively. Active C in the soil was also measured to evaluate the effects of the amendments on biological soil quality. The Pb contaminated soil was diluted in serial with uncontaminated soil for the bioassays. Seed germination and root elongation tests using lettuce (Lactuca sativa) showed increases in germination percentage and root length in soil treated with the amendments. Biochar was most effective and increased seed germination by 360% and root length by 189% compared to the unamended soil. Up to 20% soil dilution resulted in more than 50% seed germination. Bioavailability and bioaccessibility of Pb in the soils were decreased by 92.5% and 48.5% with mussel shell, by 84.8% and 34.5% with cow bone, and by 75.8% and 12.5% with biochar, respectively, compared to the unamended soil. We found that the Pb availability in the military shooting range soil can be reduced effectively by the tested amendments or soil dilution alternately, thereby decreasing the risk of ecotoxicity. The most toxic soils were organic-poor (1-10 g C kg(-1)), sandy soils (c. 74% sand), with intermediate metal concentrations (e.g. 7150-13,100 mg Pb kg(-1), 2970-53,400 mg Zn kg(-1)). Significant relationships between soil properties and the life cycle parameters were determined. The best coefficients of correlation were generally found for texture, pH, Ag, Cd, Mg, Pb, Tl, and Zn both singularly and in multivariate regressions. Glyphosate and AMPA concentrated in the first centimetre of the top soil layer and the downward transport of glyphosate and AMPA was only detected in the earthworm burrows, ranging from 0.04 to 4.25\u202f\u03bcg\u202fg-1 for glyphosate and from 0.01 (less than limit of detection) to 0.76\u202f\u03bcg\u202fg-1 for AMPA. Different soil moisture contents (expressed as % of the soil water holding capacity, WHC) were evaluated: 50% (standard soil moisture conditions for soil invertebrates' tests); 75% (to simulate increasing soil water availability after intense rainfalls and/or floods); 40%, 30%, 25% and 20% (to simulate decreasing soil water availability during droughts). Invertebrates' avoidance behavior and changes in soil porewater major ions and metal(loid)s were assessed after 48\u202fh exposure. Soil incubations induced a general solubilization/mobilization of porewater major ions, while higher soil acidity favored the solubilization/mobilization of porewater metal(loid)s, especially at 75% WHC. Folsomia candida preferred soils moistened at 50% WHC, regardless the soils were contaminated or not and the changing soil porewater characteristics. X-ray tomography showed that changes in macro porosity and pore size distribution as a result of bioturbation were not different between columns with and without Ag2S-NPs. Generally, extractable residues of BDE-47 were dominant in sediment and posed high environment risk. The amount of non-extractable residues (NERs) accounted for 39.0% of initial radioactivity in oxic sediments was significantly higher than those in anoxic sediments (17.6%). Most of NERs were localized in the humin fraction and presented as sequestrated forms. Under oxic conditions, the present of Limnodrilus significantly increased the proportion of NERs in sediment. Limnodrilus accumulated 34.2% of initial radioactivity. Under anoxic conditions, the addition of iron (\u2162) [Fe(III)], sulfate and nitrate reduced the environmental risk of BDE-47 with the increase of NERs formation, while manganese (IV) [Mn(IV)] addition had no effect on the formation of NERs. The present of Limnodrilus and electron acceptors promoted the production of metabolites. Meanwhile, BDE-47 changed the microbial community structure of sediments. Although its effects are evident in the healthcare settings, the root cause should be traced back to the One Health link, extending from animals to the environment. In fact, the use of organic fertilizers in agroecosystems represents one, if not the primary, cause of the introduction of antibiotics and antibiotic-resistant bacteria into the soil. Since the concentrations of antibiotics introduced into the soil are residual, the agroecosystem has become a perfect environment for the selection and proliferation of antibiotic resistance genes (ARGs). The continuous influx of these emerging contaminants (i.e., antibiotics) into the agroecosystem results in the selection and accumulation of ARGs in soil bacteria, occasionally giving rise to multi-resistant bacteria. These bacteria may harbour ARGs related to various antibiotics on their plasmids. In this context, these bacteria can potentially enter the human sphere when individuals consume food from contaminated agroecosystems, leading to the acquisition of multi-resistant bacteria. Once introduced into the nosocomial environment, these bacteria pose a significant threat to human health. In this review, we analyse how the use of digestate as an organic fertilizer can mitigate the spread of ARGs in agroecosystems. Furthermore, we highlight how, according to European guidelines, digestate can be considered a Nature-Based Solution (NBS). The concept that different resources co-limit plant productivity has been well studied. However, co-limitation of soil microbial communities by multiple resources has not been as thoroughly investigated. Specifically, it is not clearly understood how microbial communities respond to shifts in multiple interacting resources such as water, temperature, and nitrogen (N), in the context of global change. To test the effects of these various resources on soil microorganisms, we established a field experiment with temperature and N manipulation in three grasslands of northern China, where there is a decrease in precipitation from east to west across the region. We found that microbial responses to temperature depended upon seasonal water regimes in these temperate steppes. When there was sufficient water present, warming had positive effects on soil microorganisms, suggesting an interaction between water and increases in temperature enhanced local microbial communities. When drought or alternating wet\u2013dry stress occurred, warming had detrimental effects on soil microbial communities. Our results also provide clear evidence for serial co-limitation of microorganisms by water and N at the functional group and community levels, where water is a primary limiting factor and N addition positively affects soil microorganisms only when water is sufficient. Using a between-subjects design with information treatments, we examined the impact of informing consumers about potential human health and environmental effects linked to microplastics pollution on their valuation for the technology. We found that the information treatments increased consumers\uffe2\uff80\uff99 willingness to pay for them. Specifically, consumers\uffe2\uff80\uff99 willingness to pay for mussels with a 90% depuration efficiency certification is around US$ 4. The provision of health impact information increased the price premium by 56%, while the provision of environmental information increased it by 21%. Furthermore, combined health and environmental information significantly increased the probability of non-purchasing behavior by 22.8% and the risk perception of microplastics for human health by 5.8%. In the present study, we report the design and optimization of a dual-labelled TaqMan \u2122 probe targeting the V7 region of 16S rRNA gene for the detection of LAB in CSS. This quantitative PCR (qPCR) assays is useful for the simultaneous detection of the ten LAB genera communally encountered in CSS as Aerococcus, Carnobacterium, Enterococcus, Lactobacillus, Lactococcus, Leuconostoc, Macrococcus, Streptococcus, Vagococcus and Weissella. The specificity of this method was demonstrated against 14 genera (44 isolates, 35 species) of Gram-positive bacteria and 19 genera of Gram-negative (40 isolates, 34 species). Calibration of the method was performed in CSS matrix using a mix of equimolar cultured solution of five LAB. Quantification with the qPCR method range from 3.5 to 8.5 Log CFU/g in CSS matrix, covering 5 orders of magnitude. On these artificially contaminated CSS slices, PCR method results correlated successfully (R2\u00a0=\u00a00.9945) with the conventional enumeration on Elliker medium. In addition, the new method was successful on commercial CSS from five different origins with a quantification range from 3.7 Log CFU/g to 8.0 Log CFU/g. A total of 18 L. monocytogenes isolates were obtained from 229 samples, and their genomes were sequenced to perform comparative genomic analyses. An increase in the frequency of isolation of L. monocytogenes and in the diversity of sequence types (STs) detected was observed along time. Although the strains isolated belonged to six different STs (ST8, ST9, ST14, ST37, ST121 and ST155), ST9 was the most abundant (8 out of 18 strains). Low (0 and 2) single nucleotide polymorphism (SNP) distances were found between two pairs of ST9 strains isolated in both cases 3\u00a0months apart from the same processing room (Lm-1267 and Lm-1705, with a 2 SNPs distance in the core genome; Lm-1265 and Lm-1706, with a 0 SNPs distance), which suggests that these strains may be persistent L. monocytogenes strains in the food processing environment. Most strains showed an in silico attenuated virulence potential either through the truncation of InlA (in 67% of the isolates) or the absence of other virulence factors involved in cell adhesion or invasion. Twelve of the eighteen L. monocytogenes isolates contained a plasmid, which ranged in size from 4 to 87 Kb and harbored stress survival, in addition to heavy metals and biocides resistance determinants. Identical or highly similar plasmids were identified for various sets of L. monocytogenes ST9 isolates, which suggests the clonal expansion and persistence of plasmid-containing ST9 strains in the processing environments of the meat facility. Finally, the analysis of the L. monocytogenes genomes available in the NCBI database, and their associated metadata, evidenced that strains from ST9 are more frequently reported in Europe, linked to foods, particularly to meat and pork products, and less represented among clinical isolates than other L. monocytogenes STs. This is achieved through a case study analysis of the Kalahari rangelands in southwest Botswana. The paper first describes the socio-economic and ecological characteristics of the Kalahari rangelands and the types of land degradation taking place. It then focuses on bush encroachment as a way of exploring new economic instruments (e.g. Payments for Ecosystem Services) designed to enhance the flow of ecosystem services that support livelihoods in rangeland systems. It does this by evaluating the likely impacts of bush encroachment, one of the key forms of rangeland degradation, on a range of ecosystem services in three land tenure types (private fenced ranches, communal grazing areas and Wildlife Management Areas), before considering options for more sustainable land management in these systems. Encroachment or woody thickening, is thought to result from overgrazing, changes in fire regimes and increased atmospheric carbon dioxide concentrations. Eighteen years after one-off shrub removal by ploughing we assessed the effects of four different land management systems resulting from two levels each of grazing (grazed, ungrazed) with and without ploughing, on the cover of landscape units, soil surface condition, diversity of understorey plants and density of shrubs. We recorded 2-7 times more patches under conventional conservation (unploughed-ungrazed) than the others treatments, and plant cover and diversity were greater on the two conservation (ungrazed) plots, irrespective of ploughing. Soils under shrubs and log mounds had greater indices of infiltration, stability and nutrients. Shrub density under the active pastoral (ploughed-grazed) treatment was two and a half times greater than that in other treatments, but results were not significant. The effects of different treatments on shrubs were largely species-specific. A buffer strip experiment consisting of two tree species (Eucalyptus camaldulensis and Casuarina cunninghamiana) with two planting densities and a pasture treatment was conducted to determine the effectiveness of NO(3)-N removal from a cattle feedlot effluent disposal area at Tullimba near Armidale, NSW Australia. Different management methods were applied for the buffers where grass and weeds were mowed 2-3 times during the second and third years and were not managed during the rest experimental years for the tree buffer, while grass was harvested 1-3 times per year for the pasture buffer. The differences between tree species and planting density significantly affected tree growth, but the growth difference did not significantly affect their capacities to reduce NO(3)-N in soil surface runoff and groundwater. On average for all the tree and pasture treatments, the buffer strips reduced NO(3)-N concentration by 8.5%, 14.7% and 14.4% for the surface runoff, shallow and deep groundwater respectively. The tree and pasture buffer strips were not significantly different in NO(3)-N reduction for both shallow and deep groundwater while the pasture buffer strips reduced significantly more NO(3)-N concentration in surface runoff than the tree buffer strips. About 89% of this grassland C is stored in the soil and is affected by management activities but the effects of these management activities on C storage under different climate settings are not known. In this study, we synthesized the effects of fertilizer (nitrogen and phosphorus) application, liming and grazing regime on the stock of SOC in global grasslands, under different site specific climatic settings using a meta-analysis of 341 datasets. We found an overall significant reduction (-8.5%) in the stock of SOC in global managed grasslands, mainly attributable to grazing (-15.0%), and only partially attenuated by fertilizer addition (+6.7%) and liming (+5.8%), indicating that management to improve biomass production does not contribute sufficient organic matter to replace that lost by direct removal by animals. Management activities had the greatest effect in the tropics (-22.4%) due primarily to heavy grazing, and the least effect in the temperate zone (-4.5%). The negative management effect reduced significantly with increasing mean annual temperature and mean annual precipitation in the temperate zone, suggesting that temperate grassland soils are potential C sinks in the face of climate change. For a sustainable management of grasslands that will provide adequate forage for livestock and mitigate climate change through C sequestration, we recommend that future tropical grassland management policies should focus on reducing the intensity of grazing. Also, to verify our findings for temperate grasslands and to better inform land management policy, future research should focus on the impacts of the projected climate change on net greenhouse gas exchange and potential climate feedbacks.", "keywords": ["2. It has only very recently been adopted in Europe and is underutilized compared to other continents where this method has been successfully implemented for over a century. This is in sharp contrast to the BC of invertebrate pests, which has been taken up widely within Europe. It has been suggested that this is related to the risk-adverse attitude of Europeans towards weed BC. Scientific and public perception have a major influence on environmental policy actions. Public perception and knowledge regarding BC is an understudied subject despite its relevance for the application of this management alternative. We aimed to assess the knowledge and perception of BC of IAP among European professionals compared with their peers from other continents. To this end we conducted an online survey including multiple choice and open questions among over 700 people professionally engaged with managing the natural environment, of which approximately half were from Europe (EU) and the others from outside (non-EU). We assessed relationships between the geographical location of the respondents and their knowledge, and perceptions of BC of weeds versus BC of invertebrate pests. We found that respondents' location influenced both perceptions and knowledge of BC for weeds. Compared to non-EU respondents, EU professionals showed less appreciation for BC (e.g., regarding safety, sustainability, and cost-effectiveness), and perceived it as a riskier method, particularly in the case of practitioners and researchers. More profoundly insect pest BC tended to be considered less safe than weed BC for non-EU respondents. Confidence in weed BC as a method, as well as in the validity of the associated pre-release risk assessments, strongly increased with the level of expertise in weed BC. While a much higher proportion of non-EU respondents were correctly aware of the presence/absence of BC in their own countries and identified successful examples of BC accurately, both groups of respondents were similarly aware of unsuccessful BC examples, including BC agents against animals, stressing the bias of EU respondents towards examples of BC failure. The appreciation of weed BC in Europe could be elevated by a combination of increasing knowledge of the technique and pre-release risk assessment and promoting successful examples of weed BC, which may bring major benefits for the management of IAP across the region.", "keywords": ["2.
