This, together with an increasing demand for fertilizer and pesticide reduction has led to consider different and new approaches. We investigate the effect of Hartmannibacter diazotrophicus strain E19, a plant growth promoting rhizobacterium, in order to enhance the production of wheat and barley under low energy input. For that, we analyze the effect of three factors: organic fertilizer (with and without, only wheat), row distance (15 cm and 50 cm), and bacterial inoculation (E19 and control) at two different organic experimental field stations (Gladbacherhof and Kleinhohenheim). The dataset provides information about different parameter determined from rhizosphere soil and root samples collected at two different developmental stages (flowering and milk/fully ripe), as well as, the grain yield, straw yield, 1000 kernel mass, and seed quality during the seasons 2020-2021 and 2021-2022. The following soil parameter were evaluated: ammonium, nitrate, carbon-nitrogen ratio, microbial biomass, the respiration activity using different carbon sources (glucose, galactose, N-acetylglucosamine, arginine). These data sets are currently under evaluation and complemented with the quantification from root samples through quantitative PCR (qPCR) of strain E19 and the metabarcoding sequencing of the bacterial communities based on 16S rRNA genes obtained from rhizosphere soil samples during the two seasons. Therefore, the impact of soil water and integrated fertilization after eight years on prokaryotic microbial communities in different compartments of root zone and their association with wheat nitrogen (N) absorption and yield were investigated. The results showed that compared with fertilization treatments (F), water regimes (W) more drastically modulated the prokaryotic microbial community structure and diversity in bulk soil, rhizosphere and endosphere. The increase of irrigation improved the prokaryotic diversity in the rhizosphere and endosphere while decreased the diversity in the bulk soil. Application of organic fertilizers significantly improved soil organic matter (SOM) and nutrient contents, increased rhizosphere and endophytic prokaryotic microbial diversity, and elevated the relative abundance of aerobic ammonia oxidation and nitrification-related functional microorganisms in rhizosphere and endosphere. Increasing irrigation elevated the relative abundance of functional microorganisms related to aerobic ammonia oxidation and nitrification in the rhizosphere and endosphere. Soil water content (SWC) and NH4+-N as well as NO3\u2212-N were key predictors of prokaryotic microbial community composition under W and F treatments, respectively. Appropriate application of irrigation and organic fertilizers increased the relative abundance of some beneficial bacteria such as Flavobacterium. Water and fertilization treatments regulated the prokaryotic microbial communities of bulk soil, rhizosphere and endosphere by altering SWC and SOM, and provided evidence for the modulation of prokaryotic microorganisms to promote nitrogen uptake and wheat yield under long-term irrigation and fertilization. However, a full understanding of the general pattern of ecosystem belowground processes in response to soil acidification due to the impacting factors remains elusive. We conducted a meta-analysis of soil acidification impacts on belowground functions using 304 observations from 49 independent studies, mainly including soil cations, soil nutrient, respiration, root and microbial biomass. Our results show that acid addition significantly reduced soil pH by 0.24 on average, with less pH decrease in forest than non-forest ecosystems. The response ratio of soil pH was positively correlated with site precipitation and temperature, but negatively with initial soil pH. Soil base cations (Ca2+, Mg2+, Na+) decreased while non-base cations (Al3+, Fe3+) increased with soil acidification. Soil respiration, fine root biomass, microbial biomass carbon and nitrogen were significantly reduced by 14.7%, 19.1%, 9.6% and 12.1%, respectively, under acid addition. These indicate that soil carbon processes are sensitive to soil acidification. Overall, our meta-analysis suggests a strong negative impact of soil acidification on belowground functions, with the potential to suppress soil carbon emission. Up to 20%\uffe2\uff80\uff9330% of ASVs (particularly rare ones) were unique to individual aggregates selected within a few centimetres. Aggregates and bulk soil samples showed partly different dominant phyla, indicating that taxa that are potentially driving biogeochemical processes at the small scale may not be recognized when analysing larger soil volumes. Microbial community composition and richness of individual aggregates were closely related to aggregate-specific carbon and nitrogen content, carbon stable-isotope composition, and soil moisture, indicating that aggregates provide a stable environment for sufficient time to allow co-development of communities and their environment. We conclude that the soil microbiome is a metacommunity of variable subcommunities. Soil organic carbon (SOC) and microbial biomass carbon (MB-C) content of rhizosphere soil under both zero tillage and residue removal treatments were significantly higher than those in the bulk soil. Microbes in bulk soil presented a preferential utilization of diverse carbon sources when crop residue was retained. Zero tillage significantly increased the utilization of most carbon sources of microbial in the rhizosphere compared to conventional tillage. Principal component analysis (PCA) of the distribution of carbon substrate utilization for all treatments suggests that the microbial community catabolic diversity is different between the tillage management treatments and between soil sampling positions. To allow the crops an easier access to deep soil layers a mechanical strip-wise subsoil loosing with the addition of biocompost (DLB treatment) was established in frame of Soil3 project (for further details: Schmittmann et al. 2021). In this study, the influence of subsoil management on microbial community composition was investigated 2020 in a field trail in Klein-Altendorf (Bonn), using the three different winter wheat genotypes Milaneco, Capo and Trebelir. Thereby, exhibit the genotypes Milaneco and Capo a more deep-rooting and the genotype Trebelir a shallower rooting system. We postulate, that (i) the absolute abundance of bacteria and archaea will be increased in DLB treatment plots. And we expected that (ii) that DLB treatment will create a shift in microbial community composition, especially in the subsoil layers. Soil and roots samples were taken from 0-30, 30-60 and 60-100 cm in June 2020, at flowering time of winter wheat, in three biological replicates. The soil type was classified as Haplic Luvisol (Hypereutric, Siltic) and is characterized by a silty clay loam texture with clay accumulation in the subsoil (between 45 and 95 cm soil depth). The copy numbers of the bacterial 16S rRNA gene and archaeal 16S rRNA gene was quantified by quantitative real-time PCR (qPCR). Subsequently, 16S amplicon sequencing was performed for all bulk soil and root samples and raw sequencing data is available at the sequencing read archive (SRA) under the BioProject PRJNA1129315 (SAMN42150195, SAMN42152877).  Our data indicate a slightly increase in bacteria in soil samples among all genotypes and soil depths, this trend is not so evident for abundance of archaea in soil samples. For root samples an increase in bacterial and archaeal abundance was only observed for the genotype Capo, especially for the depths 0-30 and 30-60 cm. Alpha diversity was increased in the deep-rooting genotypes in root samples. In soil samples only for the genotype Milaneco a strong increase in alpha diversity was observed, especially in the samples from 30-60 cm.  Furthermore, beta diversity for soil and root samples was only influenced by the different sampling depths but not by the genotype or the treatment. Therefore, the impact of soil water and integrated fertilization after eight years on prokaryotic microbial communities in different compartments of root zone and their association with wheat nitrogen (N) absorption and yield were investigated. The results showed that compared with fertilization treatments (F), water regimes (W) more drastically modulated the prokaryotic microbial community structure and diversity in bulk soil, rhizosphere and endosphere. The increase of irrigation improved the prokaryotic diversity in the rhizosphere and endosphere while decreased the diversity in the bulk soil.
