The genetic regulation of strategic breeding traits, including plant architecture, growing cycle length and yield, is unknown. This study aimed to identify associations between 16 669 single nucleotide polymorphisms (SNPs) and 9 agronomic traits on a panel of 223 L. mutabilis accessions, grown in four environments, by applying a genome wide association study (GWAS). Seven environment-specific QTLs linked to vegetative yield, plant height, pods number and flowering time, were identified as major effect QTLs, being able to capture 6 to 20% of the phenotypic variation observed in these traits. Furthermore, two QTLs across environments were identified for flowering time on chromosome 8. The genes FAF, GAMYB and LNK, regulating major pathways involved in flowering and growth habit, as well as GA30X1, BIM1, Dr1, HDA15, HAT3, interacting with these pathways in response to hormonal and environmental cues, were prosed as candidate genes. These results are pivotal to accelerate the development of L. mutabilis varieties adapted to European cropping conditions by using marker-assisted selection (MAS), as well as to provide a framework for further functional studies on plant development and phenology in this species.</p", "keywords": ["0301 basic medicine", "2. Extruded and non-extruded quinoa (red and white) flour, potato starch, tara gum, and potato, pea and rice protein isolates were investigated in different recipes, some of them included egg white as an initial reference point. Results showed that extruded quinoa flour, potato starch and tara gum had deteriorating effects on GF and egg-free pasta firmness and cooking quality. Lupine flour addition itself was not able to replace egg white when added in the same amounts, but after increasing the concentration to 12%, the firmness and cooking quality decreasing effects could be improved again, especially when tara gum was absent in the formulations. In the final recipe, the content of lupine flour was increased to 30% because its protein is complementary to the quinoa protein. From the three studied protein isolates, pea protein was superior to potato or rice protein, addition of the oxidizing enzyme POx could even further improve texture firmness. However, the links between microbial diversity and ecosystem processes are not well understood. This study was designed to gain better understanding of the effects of short-term management practices on the microbial community and how changes in the microbial community affect key soil processes. The effects of different forms of nitrogen (N) on soil biology and N dynamics was determined in two soils with organic and conventional management histories that varied in soil microbial properties but had the same fertility. The soils were amended with equal amounts of N (100\u00a0kg\u00a0ha\u22121) in organic (lupin, Lupinus angustifolius L.) and mineral form (urea), respectively. Over a 91-day period, microbial biomass C and N, dehydrogenase enzyme activity, community structure of pseudomondas (sensu stricto), actinomycetes and \u03b1 proteobacteria (by denaturing gradient gel electrophoresis (DGGE) following PCR amplification of 16S rDNA fragments) and N mineralisation were measured. Lupin amendment resulted in a two- to five-fold increase in microbial biomass and enzyme activity, while these parameters did not differ significantly between the urea and control treatments. The PCR\u2013DGGE analysis showed that the addition of mineral and organic compounds had an influence on the microbial community composition in the short term (up to 10 days) but the effects were not sustained over the 91-day incubation period. Microbial community structure was strongly influenced by the presence or lack of substrate, while the type of amendment (organic or mineral) had an effect on microbial biomass size and activity. These findings show that the addition of green manures improved soil biology by increasing microbial biomass and activity irrespective of management history, that no direct relationship existed among microbial structure, enzyme activity and N mineralisation, and that microbial community structure (by PCR\u2013DGGE) was more strongly influenced by inherent soil and environmental factors than by short-term management practices.", "keywords": ["2. To address this issue we assessed: (1) the effects of the presence of the perennial grassland N(2 )fixer, Lupinus perennis, on biomass accumulation and plant N concentrations of nine-species plots of differing plant composition; (2) leaf-level physiology of co-occurring non-fixing species (Achillea millefolium, Agropyron repens, Koeleria cristata) in these assemblages with and without Lupinus; (3) the effects of elevated CO(2) on Lupinus growth and symbiotic N(2) fixation in both monoculture and the nine-species assemblages; and (4) whether assemblages containing Lupinus exhibit larger physiological and growth responses to elevated CO(2 )than those without. This study was part of a long-term grassland field experiment (BioCON) that controls atmospheric CO(2) at current ambient and elevated (560 micromol mol(-1)) concentrations using free-air CO(2) enrichment. Nine-species plots with Lupinus had 32% higher whole plot plant N concentrations and 26% higher total plant N pools than those without Lupinus, based on both above and below ground measurements. Co-occurring non-fixer leaf N concentrations increased 22% and mass-based net photosynthetic rates increased 41% in plots containing Lupinus compared to those without. With CO(2) enrichment, Lupinus monocultures accumulated 32% more biomass and increased the proportion of N derived from fixation from 44% to 57%. In nine-species assemblages, Lupinus N derived from fixation increased similarly from 43% to 54%. Although Lupinus presence enhanced photosynthetic rates and leaf N concentrations of co-occurring non-fixers, and increased overall plant N pools, Lupinus presence did not facilitate stronger photosynthetic responses of non-fixing species or larger growth responses of overall plant communities to elevated CO(2). Non-fixer leaf N concentrations declined similarly in response to elevated CO(2) with and without Lupinus present and the relationship between net photosynthesis and leaf N was not affected by Lupinus presence. Debittering of lupin seeds has a long history; however, the control of the level of alkaloids after processing the seeds is typically only evaluated by changes in the bitter taste. The aim of this study was to evaluate the profile and residual levels of quinolizidine alkaloids (QA) in (Lupinus mutabilis Sweet) after aqueous debittering process. Samples from 10 ecotypes from different areas of Peru were analyzed before and after the process. Based on results obtained by gas chromatography and mass spectrometry, from eight alkaloids identified before the debittering process, only small amounts of lupanine (avg. 0.0012\uffc2\uffa0g/100\uffc2\uffa0g DM) and sparteine (avg. 0.0014\uffc2\uffa0g/100\uffc2\uffa0g DM) remained in the seeds after the debittering process, and no other alkaloids were identified. Once the culture cycle is over, after shoot harvesting, a progressive transference of contaminants from roots to soil may take place as decomposition of roots occurs. An incubation experiment with Cu, Zn, Cd, and As-loaded roots of white lupin and soils with different pH values and concentrations of these contaminants from the area affected by a mine spill at Aznalc\u00f3llar (near Seville, Spain) was performed in order to assess the effect of the decomposition of the roots to the pH and (NH4)2SO4-extractable levels of these pollutants in the soils. Pollutants loaded-roots were mineralized (56 d) at a ratio similar to animal manures (15.8-19.4% of total organic carbon) in soil. The estimated root inputs of contaminants in comparison to their extractable concentrations in soil were high, especially in the control, non-contaminated and neutral contaminated soils. However, the extractable concentrations of the toxic elements in the soil were mainly governed by soil pH. Such compounds are involved in seed coat colour and stem pigmentation. Lupinus mutabilis (tarwi) is a legume crop domesticated in the Andean region, valued for the high protein and oil content of its seeds. Tarwi accessions are being selected for cultivation in Europe under defined breeding criteria. Seed coat colour patterns are relevant breeding traits in tarwi, and these are conditioned by anthocyanin content. We identified and isolated part of the tarwi flavonone 3-hydroxylase gene (LmF3h) from two accessions with distinct seed coat colour patterns. Two partial LmF3h paralogues, with predicted 20% amino-acid changes but little predicted tertiary structure alterations, were identified in the coloured seed genotype, while only one was present in the white seed genotype. Upon selection and validation of appropriate reference genes, a RT-qPCR analysis showed that these paralogues have different levels of expression during seed development in both genotypes, although they follow the same expression patterns. DNA and transcription analyses enabled to highlight potential F3H paralogues relatable to seed coat pigmentation in tarwi and, upon biochemical and genetic confirmation, prompt marker-assisted breeding for relevant phenotypic traits associated with flavonoid synthesis.</p></article>", "keywords": ["0301 basic medicine", "2. Yellow lupines accumulated heavy metals mainly in roots (Cu, Cd and especially Pb were poorly translocated to shoots). This indicates a potential use of this plant in metal phytostabilisation. Starting with a classic histological approach, we described rootlet primordium development and defined eight developmental stages from rootlet initiation to their emergence. We observed the establishment of an auxin maximum through rootlet development in transgenic roots expressing the DR5:GUS auxin reporter. All tarwi accessions were classified as susceptible, exhibiting sporulating necroses on the main stem from seven days after inoculation. Anthracnose severity was lower on anthocyanin-rich tarwi plants, with accession LM34 standing out as the less susceptible. Accession I82 better combines anthracnose response and yield. In global terms, disease severity was lower on white lupin than on tarwi. This study aimed to identify associations between 16 669 single nucleotide polymorphisms (SNPs) and 9 agronomic traits on a panel of 223 L. mutabilis accessions, grown in four environments, by applying a genome wide association study (GWAS). Seven environment-specific QTLs linked to vegetative yield, plant height, pods number and flowering time, were identified as major effect QTLs, being able to capture 6 to 20% of the phenotypic variation observed in these traits. Furthermore, two QTLs across environments were identified for flowering time on chromosome 8. The genes FAF, GAMYB and LNK, regulating major pathways involved in flowering and growth habit, as well as GA30X1, BIM1, Dr1, HDA15, HAT3, interacting with these pathways in response to hormonal and environmental cues, were prosed as candidate genes. Principal component analyses pointed out flowering time, yield, and architecture-related traits as the main factors explaining variation between accessions. The Peruvian material stands out among the top-yielding accessions in Europe, characterized by early lines with high grain yield (e.g., LIB065, LIB072, and LIB155). Bolivian and Ecuadorian materials appear more valuable for the selection of genotypes for Andean conditions and for biomass production in Europe. We also observed that flowering time in the different environments is influenced by temperature accumulation. Within the panel, it is possible to identify both early and late genotypes, characterized by different thermal thresholds (600\uffc2\uffb0C\uffe2\uff80\uff93700\uffc2\uffb0C and 1,000\uffe2\uff80\uff931,200\uffc2\uffb0C GDD, respectively). Combined Analysis over Location presented a significant accession-location interaction for traits of economic interest such as seed crude protein and 100 seed weight. Seed crude protein was higher in L. mutabilis accessions (up to 43.8 g 100 g\uffe2\uff88\uff921 seed) than white and blue lupins. Floral traits such as color and size are related to pollinators diversity and preference. Insect pollinators and floral traits interplay among different lupin species was examined. Pollinators visited L. mutabilis flowers belong to the genera Anthophora, Bombus, Megachile and Xylocopa. The beneficial interaction of L. mutabilis with Bradyrhizobium strains in the soil and various pollinator species is also discussed, and the effect of abiotic stresses on these interactions is highlighted. Prospects of alternative uses of L. mutabilis biomass in Northern Europe and opportunities for breeding strategies are discussed. Seed coat colour patterns are relevant breeding traits in tarwi, and these are conditioned by anthocyanin content. We identified and isolated part of the tarwi flavonone 3-hydroxylase gene (LmF3h) from two accessions with distinct seed coat colour patterns. Two partial LmF3h paralogues, with predicted 20% amino-acid changes but little predicted tertiary structure alterations, were identified in the coloured seed genotype, while only one was present in the white seed genotype. Upon selection and validation of appropriate reference genes, a RT-qPCR analysis showed that these paralogues have different levels of expression during seed development in both genotypes, although they follow the same expression patterns. Despite its high nutritional value and promotion efforts by regional researchers and breeders, tarwi is not a widely cultivated crop in its center of origin. In this review, we present the work carried out by public breeding programs of L. mutabilis at national agricultural research institutes, universities, and other institutions in Ecuador, Peru, and Bolivia. The main breeding method used in the Andes to improve local landraces has been mass selection to adapt lines to specific environments. At least 25 cultivars or ecotypes have been selected and released over the last 40 years using this breeding system. Nevertheless, breeders are currently struggling to develop new varieties that are high yielding, suitable for mechanized harvesting, have a low content of alkaloids or other anti-nutritional properties, and resistant to anthracnose (Colletotrichum acutatum). Therefore, it is necessary to reassess the potential of this crop and invest in its research to incorporate new techniques and breeding strategies to optimize the development of new varieties in the Andes which address the current cultivation challenges of the species.</p></article>", "keywords": ["2. Within the panel, it is possible to identify both early and late genotypes, characterized by different thermal thresholds (600\uffc2\uffb0C\uffe2\uff80\uff93700\uffc2\uffb0C and 1,000\uffe2\uff80\uff931,200\uffc2\uffb0C GDD, respectively). Indications on top-yielding and early/late accessions, heritability of morpho-physiological traits, and their associations with grain yield are reported and remain largely environmental specific, underlining the importance of selecting useful genetic resources for specific environments. The current study aims to evaluate the performance of six selected L. mutabilis accessions in a Mediterranean area using two different planting dates for two consecutive experimental years. A split plot design with accessions as subplots and planting dates as main plots was used. Measurements such as days to flowering, plant height, total number of pods and seeds per plant, seed yield and seed crude protein content were taken. All accessions performed better when cultivation started not later than the end of autumn since high temperatures occurred during late spring shortened the growth cycle and reduced yield. Earlier cultivation applied, in the middle of autumn, did not enhance significantly seed yield production. For this purpose, eight Andean lupin accessions, one white and one blue lupin commercial varieties were evaluated for 71 agro-morphological traits in a Randomized Complete Block design with three replications. Combined Analysis over Location presented a significant accession-location interaction for traits of economic interest such as seed crude protein and 100 seed weight. Seed crude protein was higher in L. mutabilis accessions (up to 43.8 g 100 g\u22121 seed) than white and blue lupins. Andean lupin yielded up to 327 kg ha\u22121 (LIB214) in Kalamata, while its yield was lower than the white lupin in Athens. Using principal component analysis, three groups of accessions were formed, one by each lupin species and three within Andean lupin accessions. Floral traits such as color and size are related to pollinators diversity and preference. Insect pollinators and floral traits interplay among different lupin species was examined. Pollinators visited L. mutabilis flowers belong to the genera Anthophora, Bombus, Megachile and Xylocopa. AMF come from soil or root samples from a two growing-season mesocosm experiment mimicking a crop rotation, with a first phase growing AM or non-AM and/or rhizobial or non rhizobial precrops followed with an AM crop. During the second phase, organic fertilization was applied (saw dust or wheat straw). The aim is to test the effect of precrop plant symbiotic functional group on following crop AM fungal diversity and abundance. Four crop plant species were used to have a combination of all symbiotic groups: spring canola (Brassica napus cv. Medicus, NPZ, non-rhizobial / non-AM), withe lupine (Lupinus albus cv. Energy, Feldsaaten Freudenberger, rhizobial / non-AM),  spring barley (Hordeum vulgare cv. Barke, Saatzucht Breun, non rhizobial/AM) faba bean (Vicia faba cv. Tiffany, NPZ, rhizobial / AM). The succeeding crop was winter barley (Hordeum vulgare, cv. Antonella, Nordsaat Saatzucht, non-rhizobial / AM). The AMF community structure was analyzed by amplicon sequencing of the D1-D2 region of the fungal LSU (28S) rRNA gene using the primers LR2rev (5'-GAAAAGAACTTTGAAAA-3') and LR3 (5'-CCGTGTTTCAAGACG-3'). The taxonomy and sequence for each sequence variant is included. The sequences were generated using the primers LR2rev (5'-GAAAAGAACTTTGAAAA-3') and LR3 (5'-CCGTGTTTCAAGACG-3'). The results revealed that the mycorrhizal precrop symbiotic group (AM or not AM), but not the rhizobial one (N2-fixer or not N2-fixer), affected the richness and composition of AM fungi in the soil. The effect on composition, but not on richness, persisted in the following crop, demonstrating a legacy effect of precrop functional group. However, this effect was weaker than that of contemporary factors such as nutrient addition. AMF community composition correlated to decreasing crop yield after AM precrops. Phylogenetic analyses revealed that differences in AMF communities pertained at deep phylogenetic levels. These findings suggest negative plant-soil feedbacks within the AM symbiosis in agro-ecosystems typical of western Europe, driven by the consistent selection of deep clades of AM fungi. In this review, abiotic and biotic constraints to L. mutabilis cultivation in European soil and climatic conditions are discussed, and cultivation management practices are suggested. The beneficial interaction of L. mutabilis with Bradyrhizobium strains in the soil and various pollinator species is also discussed, and the effect of abiotic stresses on these interactions is highlighted. Prospects of alternative uses of L. mutabilis biomass in Northern Europe and opportunities for breeding strategies are discussed. In conclusion, the different approach to crop modeling for Southern and Northern European climatic conditions is highlighted.
