Avhandlingens \u00f6vergripande fr\u00e5gest\u00e4llningar fokuserar p\u00e5: diskurser om m\u00e4ns v\u00e5ld i Socialstyrelsens utbildningsmaterial, socialarbetares kunskapsbehov och hur dessa tillgodoses, samt socialarbetares uppfattningar om m\u00e4ns v\u00e5ld mot kvinnor. Avhandlingen har ett feministiskt och intersektionellt perspektiv och bygger p\u00e5 tv\u00e5 delstudier. I den f\u00f6rsta delstudien anv\u00e4nds kvalitativa metoder, och empirin best\u00e5r av fem utbildningsmaterial om m\u00e4ns v\u00e5ld mot kvinnor som publicerats av Socialstyrelsen. Empirin analyserats med hj\u00e4lp av Carol Bacchis policy approach What\u2019s the Problem Represented to be?, och Norman Faircloughs kritiska diskursanalys. Empirin i den andra delstudien utg\u00f6rs av enk\u00e4tdata som inh\u00e4mtats bland 153 socialarbetare verksamma i socialtj\u00e4nsten i tre svenska kommuner med olika demografi och geografisk placering. Avhandlingen visar tre centrala resultat. F\u00f6r det f\u00f6rsta framst\u00e4lls m\u00e4ns v\u00e5ld i k\u00f6nsneutrala termer och v\u00e5ldet beskrivs som ett relationsv\u00e5ld och/eller familjev\u00e5ld. F\u00f6r det andra problematiserar utbildningsmaterialens texter inte m\u00e4ns \u00f6verordning, de f\u00e4ster inte heller n\u00e5gon s\u00e4rskild vikt vid strukturella orsakssammanhang. F\u00f6r det tredje centreras kunskapen och diskurserna kring individuella behov och orsaker. Fr\u00e4mst fokuseras behoven hos de grupper av kvinnor som Regeringen menar \u00e4r s\u00e4rskilt s\u00e5rbara f\u00f6r v\u00e5ld. Det senare leder till att kvinnor kollektiviseras och att m\u00e4ns ansvar f\u00f6r v\u00e5ldet, i synnerhet svenska m\u00e4n, och sambandet mellan v\u00e5ld, j\u00e4mlikhet och andra sociala strukturer f\u00f6rbises. Professionell erfarenhet i \u00e4renden d\u00e4r v\u00e5ld f\u00f6rekommer och utbildning inom omr\u00e5det \u00e4r faktorer som socialarbetarna anser bidragit till att \u00f6ka deras kunskap. Trots detta uppger respondenter med l\u00e5ng arbetslivserfarenhet, h\u00f6g utbildning, och fortbildning att de saknar kunskap om m\u00e4ns v\u00e5ld mot kvinnor. \u00c4ven fast att socialarbetarna upplever kunskapsbrist, visar resultatet att socialarbetarna endast tagit del st\u00f6ddokument i begr\u00e4nsad omfattning. Den h\u00e4r avhandlingen bidrar med kunskap om vikten av spr\u00e5kanv\u00e4ndning och hur vi f\u00f6rst\u00e5r m\u00e4ns v\u00e5ld mot kvinnor. An experiment manipulating densities of the grasshopper Oedaleus asiaticus was performed to assess top-down effects of grasshoppers on C:N:P stoichiometry of plants and soil in a grassland ecosystem in Inner Mongolia (China). With increased grasshopper feeding, plant biomass declined fourfold, litter abundance increased 30%, and the plant community became dominated by non-host plant taxa. Plant stoichiometric response depended on whether or not the plant was a grasshopper host food species: C:N and C:P ratios increased with increasing grasshopper density (GD) for host plants but decreased in non-host plants. These data suggest either a direct transfer of grasshopper-recycled nutrients from host to non-host plants or a release of non-host plants from nutrient competition with heavily grazed host plants. Litterfall C:N and C:P decreased across moderate levels of grasshopper density but no effects on C:N:P stoichiometry in the surface soil were observed, possibly due to the short experimental period. Our observations of divergent C:N:P stoichiometric response among plant species highlight the important role of grasshopper herbivory in regulating plant community structure and nutrient cycling in grassland ecosystems.", "keywords": ["Male", "2. We described change in hemoglobin, change in PSA, enucleated prostate weight, enucleation and morcellation times, length of stay, and postoperative retention rate.The average age was 71.4\u00a0years (range 53-87). Urinary retention requiring catheterization was present in seven patients (78%). Two patients had a known diagnosis of prostate cancer preoperatively. The mean preoperative PSA on average was 21.6\u00a0ng/dL. Patients had a wide range of gland sizes, with a mean enucleated weight of 141\u00a0g (range 18-344\u00a0g). Mean reduction in hemoglobin was 16.9% following HoLEP. All patients managed to void postoperatively. All but one patient went home on postoperative day 1, and this patient went home on postoperative day 2. Lead, cadmium, copper, zinc, manganese, and iron were analyzed using atomic absorption spectrometry. Statistically significant differences between plots were found for lead, cadmium, and iron in ground beetles. Correlations between ground beetles and soil or leaf litter were positive for lead and cadmium concentrations and negative for iron concentration. Differences in species metal concentrations were recorded. Higher concentrations of all studied metals were found in female beetles. However, a significant difference between sexes was found only for manganese. Given the critical role of seafood in food security, growing concerns about microplastics might be detrimental to meeting future global food demand. This study employs a discrete choice experiment to investigate Chilean consumers' preferences for technology aimed at mitigating microplastic levels in mussels. Using a between-subjects design with information treatments, we examined the impact of informing consumers about potential human health and environmental effects linked to microplastics pollution on their valuation for the technology. We found that the information treatments increased consumers\uffe2\uff80\uff99 willingness to pay for them. Specifically, consumers\uffe2\uff80\uff99 willingness to pay for mussels with a 90% depuration efficiency certification is around US$ 4. The provision of health impact information increased the price premium by 56%, while the provision of environmental information increased it by 21%. Furthermore, combined health and environmental information significantly increased the probability of non-purchasing behavior by 22.8% and the risk perception of microplastics for human health by 5.8%. The limited availability of effective drugs on the market and the increased risk of resistance development due to extensive usage, highlight the urgent need for new antischistosomal drugs. Recent studies have shown that robenidine derivatives, containing an aminoguanidine core, exhibit promising activities against Plasmodium falciparum, motivating further investigation into their efficacy against Schistosoma mansoni, due to their similar habitat and the resulting related cellular mechanisms like the heme detoxification pathway. The conducted phenotypic screening of robenidine and 80 derivatives against newly transformed schistosomula and adult Schistosoma mansoni yielded 11 candidates with low EC50 values for newly transformed schistosomula (1.12-4.63\u00a0\u03bcM) and adults (2.78-9.47\u00a0\u03bcM). The structure-activity relationship revealed that electron-withdrawing groups at the phenyl moiety, as well as the presence of methyl groups adjacent to the guanidine moiety, enhanced the activity of derivatives against both stages of Schistosoma mansoni. The two compounds 2,2'-Bis[(3-cyano-4-fluorophenyl)methylene] carbonimidic Dihydrazide Hydrochloride (1) and 2,2'-Bis[(4-difluoromethoxyphenyl) ethylidene] carbonimidic Dihydrazide Hydrochloride (19), were selected for an in vivo study in Schistosoma mansoni-infected mice based on their potency, cytotoxicity, pharmacokinetic-, and physicochemical properties, but failed to reduce the worm burden significantly (worm burden reduction <20%). They died suddenly in 1494 and their deaths have been for centuries a subject of debate. The exhumation of their remains offered the opportunity to study the cause of their death through a multidisciplinary research project. Anthropological analyses, together with documentary evidences, radiocarbon dating and ancient DNA analysis supported the identification of the remains attributed to Pico. Macroscopic examination did not reveal paleopathological lesions or signs related to syphilis. Heavy metals analysis, carried out on bones and mummified tissues, showed that in Pico's remains there were potentially lethal levels of arsenic, supporting the philosopher's poisoning theory reported by documentary sources. Pooled digestive juice extracts of four (SISE) or two (PFM) animals were used per measurement run. In both cases, digestive juice extracts in 100 \u03bcL of deionized water were sufficient for one measurement run. The total surfactant concentration of P. scaber digestive juice was determined to be 9.2 \u00b1 3.5mM and the CMC was approximately 90 \u03bcM. Despite its simplicity and cost-effectiveness, the Kato-Katz method faces challenges, including reader subjectivity and reduced sensitivity. Real-time polymerase chain reaction (qPCR) technology offers standardized readouts and higher sensitivity, making it suitable for STH diagnosis and monitoring the treatment efficacy of emodepside within the framework of randomized controlled trials.</p>   Methodology/Principal findings <p>We evaluated the performance of Kato-Katz versus qPCR for assessing treatment efficacy in terms of cure rates, of single doses of 5, 10, 15, 20, 25 and 30\uffe2\uff80\uff89mg of emodepside compared to 400\uffe2\uff80\uff89mg albendazole. Spearman\uffe2\uff80\uff99s rank correlation coefficient examined the correlation between STH eggs per gram in stool samples and qPCR Ct values. Diagnostic sensitivity of qPCR was calculated using a Bayesian latent class modelling approach with data from Ascaris lumbricoides infections. Agreement between Kato-Katz and qPCR at baseline was 93.57% for Trichuris trichiura, and 73.49% for both hookworm and A. lumbricoides. For the latter helminth qPCR demonstrated higher sensitivity (85.00% vs. 47.70%) and slightly lower specificity (93.40% vs. 99.40%) compared to Kato-Katz. We observed a fair to moderate agreement with negative correlation between Ct values and Kato-Katz egg counts. Treatment efficacy, as assessed by qPCR, was lower for all doses of emodepside and albendazole compared to Kato-Katz. Nonetheless, emodepside demonstrated higher cure rates against T. trichiura and A. lumbricoides infections compared to albendazole.</p>   Conclusion/ Significance <p>Our study confirmed that qPCR is a sensitive diagnostic method for diagnosing STH infections compared to Kato-Katz and serves as a valuable tool for determining treatment efficacy in clinical trials. We engineered diatomic dielectric metasurfaces supporting bound states in the continuum that allows high-quality resonances with accessible near-fields by in-plane symmetry breaking. The large-area metasurface chips are configured as microarrays and integrated with microfluidics on an imaging platform for real-time detection of breast cancer extracellular vesicles encompassing exosomes. The optofluidic system has high sensing performance with nearly 70 1/RIU figure-of-merit enabling detection of on average 0.41 nanoparticle/\uffc2\uffb5m2 and real-time measurements of extracellular vesicles binding from down to 204 femtomolar solutions. Interestingly, infants display greater plasmid diversity compared to mothers and other healthy adults. We find that Bacteroidota, a major bacterial phylum, serves as the primary host for gut plasmids and plays a dominant role in gut plasmid transfer events. Additionally, plasmids broaden the genetic capabilities of bacteria, with their influence on bacterial function becoming more apparent as children's gut microbiomes develop. Comparisons were made with the unsupplemented basal diets (4/2 g Ca/kg DM). The 14:0 supplementation decreased (P&lt; 0/001) total tract CH4release depending on basal diet type (interaction,P&lt; 0/001) and dietary Ca level (P&lt; 0/05,post hoctest). In the concentrate-based diet, 14:0 suppressed CH4emission by 58 and 47% with 4/2 and 9/0 g Ca/kg DM, respectively. The 14:0 effect was lower (22%) in the forage-based diet and became insignificant with additional Ca. Myristic acid inhibited (P&lt; 0/05) rumen archaea without significantly altering proportions of individual methanogen orders. Ciliate protozoa concentration was decreased (P&lt; 0/05,post hoctest) by 14:0 only in combination with 9/0 g Ca/kg DM. Rumen fluid NH3concentration and acetate:pro-pionate were decreased (P&lt; 0/05) and water consumption was lower (P&lt; 0/01) with 14:0. The use of 14:0 had no clear effects on total tract organic matter and fibre digestion; this further illustrates that the suppressed methanogenesis resulted from direct effects against methanogens. Estimates were also made of volatile fatty acid (VFA) production in the rumen at each level of feeding. In all studies three to four animals were used at each level of intake.</p><p>2. Production of VFA and of methane were both related to digestible energy (de) intake. Regression lines for both VFA production and methane productionv.deintake had significant intercepts indicating an input of endogenous, fermentable organic matter into the rumen in excess of 50 g/d.</p><p>3. The values obtained for rates of methane production were compared with those calculated from stoicheiometric equations relating rates of methane and VFA production. Comparisons of methane production with that predicted fromdeintake were also made.</p><p>4. The objective of this study was to investigate the effect of divergently ranking beef cattle for residual methane emissions (RME) on animal productivity, enteric emissions, and rumen fermentation. Dry matter intake (DMI), growth, feed efficiency, carcass output, and enteric emissions (GreenFeed emissions monitoring system) were recorded on 294 crossbred beef cattle (steers = 135 and heifers = 159; mean age 441 d (SD = 49); initial body weight (BW) of 476 kg (SD = 67)) at the Irish national beef cattle performance test center. Animals were offered a total mixed ration (77% concentrate and 23% forage; 12.6 MJ ME/kg of DM and 12% CP) ad libitum with emissions estimated for 21 d over a mean feed intake measurement period of 91 d. Animals had a mean daily methane emissions (DME) of 229.18 g/d (SD = 45.96), methane yield (MY) of 22.07 g/kg of DMI (SD = 4.06), methane intensity (MI) 0.70 g/kg of carcass weight (SD = 0.15), and RME 0.00 g/d (SD = 0.34). RME was computed as the residuals from a multiple regression model regressing DME on DMI and BW (R2 = 0.45). Animals were ranked into three groups namely high RME (&amp;gt;0.5 SD above the mean), medium RME (\uffc2\uffb10.5 SD above/below the mean), and low RME (&amp;gt;0.5 SD below the mean). Low RME animals produced 17.6% and 30.4% less (P &amp;lt; 0.05) DME compared with medium and high RME animals, respectively. A ~30% reduction in MY and MI was detected in low versus high RME animals. Positive correlations were apparent among all methane traits with RME most highly associated with (r = 0.86) DME. MY and MI were correlated (P &amp;lt; 0.05) with DMI, growth, feed efficiency, and carcass output. High RME had lower (P &amp;lt; 0.05) ruminal propionate compared with low RME animals and increased (P &amp;lt; 0.05) butyrate compared with medium and low RME animals. To address these issues, we propose a novel parameter grouping approach, which reduces the parametric space by co-estimating groups of parameters across compartments. Grouping of parameters is performed using genetic algorithms and is fully automated, based on a novel goodness-of-fit metric. To illustrate the practical application of the proposed methodology, two case studies were conducted. The first case study demonstrates the development of a new PBK model, while the second focuses on model refinement. In the first case study, a PBK model was developed to elucidate the biodistribution of titanium dioxide (TiO2) nanoparticles in rats following intravenous injection. A variety of parameter estimation schemes were employed. Comparative analysis based on goodness-of-fit metrics demonstrated that the proposed methodology yields models that outperform standard estimation approaches, while utilizing a reduced number of parameters. Despite a long domestication history in the Mediterranean basin, modern breeding efforts have been fairly scarce. Recent sequencing of its genome has provided tools for further description of genetic resources but detailed characterization of genomic diversity is still missing. Here, we report the genome sequencing of 39 accessions that were used to establish a white lupin pangenome. We defined 32\uffe2\uff80\uff89068 core genes that are present in all individuals and 14\uffe2\uff80\uff89822 that are absent in some and may represent a gene pool for breeding for improved productivity, grain quality, and stress adaptation. We used this new pangenome resource to identify candidate genes for alkaloid synthesis, a key grain quality trait. The white lupin pangenome provides a novel genetic resource to better understand how domestication has shaped the genomic variability within this crop. We then investigated whether the gut microbiome mediates part of the benefits of vegetable intake on lymphocyte counts.</p>                                Results                 <p>Higher levels of white blood cells, lymphocytes and basophils were all significantly correlated with lower habitual intake of vegetables, with vegetable intake explaining between 3.59 and 6.58% of variation in white blood cells after adjusting for covariates and multiple testing using false discovery rate (q\uffe2\uff80\uff89&lt;\uffe2\uff80\uff890.1). No such association was seen with fruit intake. A mediation analysis found that 20.00% of the effect of vegetable intake on lymphocyte counts was mediated by one bacterial genus, Collinsella, known to increase with the intake of processed foods and previously associated with fatty liver disease. Here, we explored women\u2019s participation in ecology conferences by analyzing female representation, behavior, and personal experience at the 1st Meeting of the Iberian Society of Ecology (SIBECOL). The conference had 722 attendees, 576 contributions, and 27 scientific sessions. The gender of attendees and presenters was balanced (48/52% women/men), yet only 29% of the contributions had a woman as last author. Moreover, men presented most of the keynote talks (67%) and convened most of the sessions. Our results also showed that only 32% of the questions were asked by women, yet the number of questions raised by women increased when the speaker or the convener was a woman. Finally, the post-conference survey revealed that attendees had a good experience and did not perceive the event as a threatening context for women. Yet, differences in the responses between genders suggest that women tended to have a worse experience than their male counterparts. Although our results showed clear gender biases, most of the participants of the conference failed to detect it. Twenty lactating Holstein-Friesian dairy cows (33.2\u00b16.0 kg of milk/d; 104\u00b158 d in milk at the start of the experiment) were fed a total mixed ration (corn silage-based; forage to concentrate ratio 66:34), containing either a dietary urea or a dietary nitrate source [21 g of nitrate/kg of dry matter (DM)] during 4 successive 24-d periods, to assess the methane-mitigating potential of dietary nitrate and its persistency. The study was conducted as paired comparisons in a randomized design with repeated measurements. Cows were blocked by parity, lactation stage, and milk production at the start of the experiment. A 4-wk adaptation period allowed the rumen microbes to adapt to dietary urea and nitrate. Diets were isoenergetic and isonitrogenous. Methane production, energy balance, and diet digestibility were measured in open-circuit indirect calorimetry chambers. Cows were limit-fed during measurements. Nitrate persistently decreased methane production by 16%, whether expressed in grams per day, grams per kilogram of dry matter intake (DMI), or as percentage of gross energy intake, which was sustained for the full experimental period (mean 368 vs. 310\u00b112.5 g/d; 19.4 vs. 16.2\u00b10.47 g/kg of DMI; 5.9 vs.4.9\u00b10.15% of gross energy intake for urea vs. nitrate, respectively). This decrease was smaller than the stoichiometrical methane mitigation potential of nitrate (full potential=28% methane reduction). The decreased energy loss from methane resulted in an improved conversion of dietary energy intake into metabolizable energy (57.3 vs. 58.6\u00b10.70%, urea vs. nitrate, respectively). Despite this, milk energy output or energy retention was not affected by dietary nitrate. Nitrate did not affect milk yield or apparent digestibility of crude fat, neutral detergent fiber, and starch. Our study focused on measuring methane and carbon dioxide emissions, total-tract digestibility, and ruminal fermentation in growing beef cattle fed a diet supplemented with various additives or ingredients. Two experiments, each designed as a 4 x 4 Latin square with 21-d periods, were conducted using 16 Holstein steers (initial BW 311.6 +/- 12.3 kg). In Exp. 1, treatments were control (no additive), monensin (Rumensin, Elanco Animal Health, Indianapolis, IN; 33 mg/kg DM), sunflower oil (400 g/d, approximately 5% of DMI), and proteolytic enzyme (Protex 6-L, Genencor Int., Inc., CA; 1 mL/kg DM). In Exp. 2, treatments were control (no additive), Procreatin-7 yeast (Prince Agri Products, Inc., Quincy, IL; 4 g/d), Levucell SC yeast (Lallemand, Inc., Rexdale, Ontario, Canada; 1 g/d), and fumaric acid (Bartek Ingredients Inc., Stoney Creek, Ontario, Canada; 80 g/d). The basal diet consisted of 75% barley silage, 19% steam-rolled barley grain, and 6% supplement (DM basis). Forage harvested daily from the previously grazed pastures was consumed ad libitum. Crude protein concentration was 10.3 and 13.0%, IVDMD was 64.5 and 75.3%, and the level of condensed tannins was 17.7 and 0.5% for sericea lespedeza and crabgrass/tall fescue, respectively. Dry matter intake (1.11 vs. 0.67 kg/d) and digestible DMI (estimated from IVDMD; 0.71 vs. 0.51 kg/d) were greater (P < 0.01) for sericea lespedeza than for crabgrass/tall fescue. Despite higher DMI by goats fed sericea lespedeza, daily energy expenditure (409 vs. 431 kJ/kg BW(0.75)), heart rate (70 vs. 73 beats/min), and the ratio of energy expenditure to heart rate (5.82 vs. 5.94) did not differ between sericea lespedeza and crabgrass/tall fescue, respectively (P > 0.13). Methane emission expressed as both quantity per day or relative to DMI was lower (P <0.001) for sericea lespedeza than for crabgrass/tall fescue (7.4 vs. 10.6 g/d and 6.9 vs. 16.2 g/kg DMI). At high feed intake levels, the proportion of energy lost as CH(4) decreases as the digestibility of the diet increases. Therefore, improving forage digestibility may improve productivity as DM and energy intake are increased. A zero-grazing experiment was conducted to determine the effect of sward DM digestibility (DMD) on DMI, CH(4) emissions, and indices of rumen fermentation of beef animals. Twelve Charolais-cross heifers were assigned to 1 of 2 treatments, with 6 heifers per dietary treatment. Additionally, 4 cannulated Aberdeen Angus-cross steers were randomly allocated to each of these 2 treatments in a crossover design. Dietary treatments consisted of swards managed to produce (i) high digestibility pasture (high DMD) or (ii) pasture with less digestibility (low DMD), both offered for ad libitum intake. All animals were zero-grazed and offered freshly cut herbage twice daily. In vitro DMD values for the high and low DMD swards were 816 and 706 g/kg of DM. Heifers offered the high DMD grass had greater (P < 0.001) daily DMI of 7.66 kg compared with 5.38 kg for those offered the low DMD grass. Heifers offered the high DMD grass had greater (P = 0.003) daily CH(4) production (193 g of CH(4)/d) than those offered the low DMD grass (138 g of CH(4)/d). However, when corrected for DMI, digestible DMI, or ingested gross energy, there was no difference (P > 0.05) in CH(4) production between dietary treatments. For cannulated steers, intake tended (P = 0.06) to be greater for the high DMD grass (5.56 vs. 4.27 kg of DM/d), but rumen protozoa (4.95 x 10(4)/mL; P = 0.62); rumen ammonia (34 mg of N/L; P = 0.24); rumen total VFA (103 mM; P = 0.58), and rumen pH (6.8; P = 0.43) did not differ between treatments. Saponins were then administered intraruminally in two equal doses daily into four ruminally and duodenally cannulated wethers (60 +/- 1 kg) in amounts equivalent to 0, 200, 400, and 800 mg/kg BW daily in a 4 x 4 Latin Square arrangement. Saponins increased VFA concentrations (P = .001) and lowered ruminal pH (P = .001) after 2 d of administration. On d 2 and d 14, saponins decreased (P < .01) protozoal populations. On d 11, pressure changes during ruminal contractions increased by 79% during resting (P = .06), 202% during feeding (P = .05), and 53% during an 8-h period (P = .08) as the level of saponins increased from 0 to 800 mg/kg BW. Administered saponins increased duodenal flows of OM and total N by 19 and 26%, respectively (P = .04 and .01). Apparent total tract N digestibility was reduced (P = .02) by 5% at the highest saponin dosage. We conclude that alfalfa may contain enough saponins to adversely impact ruminoreticular motility. There was no effect of tallow level on OM intake (1,103.1+/-51 g/d), OM digestibility (84+/-0.9%), GE digestibility (83+/-1.1%), or cell solubles digestibility (84.2+/-1.2%). The level of tallow quadratically decreased ADF digestibility (P < 0.05), methane emissions, and methane energy as a percentage of GE P < 0.01). There were linear increases in dietary GE (megacalories per kilogram of OM [P < 0.01]), dietary DE (megacalories per kilogram of OM [P < 0.05]), and dietary ME (megacalories per kilogram of OM [P < 0.01]), as dietary tallow increased. Numbers of ruminal protozoa (Entodinium spp. and Polyplastron sp.) decreased linearly (P < 0.05) with increased level of tallow. The energy value of tallow (calculated by difference) was low. The total-tract fatty acid digestibility of tallow was calculated by linear regression, without intercept, after accounting for the fatty acids digested from the base diet (0% tallow fed to a wether in a period). Fatty acids of the same carbon length were pooled for the regression analysis. All linear regressions were significant (P < 0.10) indicating no effect of tallow level on fatty acid digestibility. Lauric acid had low digestibility. The high digestibility of all C16 (89%) and C18 (104%) fatty acids suggests an effect of tallow on endogenous and microbial fatty acid excretion. The CD-HR was added at various concentrations (0, 0.17, 0.85 and 1.7 g/L). Treatment affected neither the pH of the medium nor the number of protozoa. Total VFA increased in a linear manner (P = 0.02), and NH3-N decreased quadratically (P = 0.04) as the concentration of CD-HR increased from 0.17 g/L to 1.7 g/L. Molar proportions of acetate decreased in a linear manner (P = 0.03), and propionate increased linearly (P = 0.008) with increasing concentrations of CD-HR. Production of methane was inhibited up to 90%, whereas accumulation of dihydrogen was increased 36-fold by 1.7 g/L of CD-HR supplementation relative to controls. The effect of CD-HR on methane production, ruminal fermentation and microbes, and digestibility was further investigated in vivo using four Holstein steers in a crossover design. The CD-HR supplement was mixed into the concentrate portion of a (1.5:1) Sudangrass hay plus concentrate mixture that was fed twice daily to the steers. Ruminal samples were collected 0, 2, and 5 h after the morning feeding. No effects of CD-HR supplementation on ruminal pH (P = 0.63) or protozoal numbers (P = 0.44) were observed. Molar proportion of acetate was decreased (P = 0.04) and propionate was increased (P = 0.005) by CD-HR treatment. Molar proportion of butyrate was increased (P = 0.05) in CD-HR-supplemented steers. Ruminal NH3-N was decreased (P = 0.05) by treatment. Blood plasma glucose concentration was increased (P = 0.02) and urea-N was decreased (P = 0.04) with CD-HR supplementation. Daily DMI was decreased (P = 0.04), and apparent digestibility of DM (P = 0.13), NDF (P = 0.14), and CP tended (P = 0.14) to be increased by treatment. Methane production was decreased (P = 0.03) by 19%, and the number of methanogens was also decreased (P = 0.03). Although N retention (P = 0.11), total viable bacteria (P = 0.15), and sulfate-reducing bacteria (P = 0.17) were not significantly altered by treatment, tendencies for increases were noted with CD-HR supplementation. The number of cellulolytic (P = 0.38) and acetogenic bacteria (P = 0.32) remained unchanged by treatment. Short- and long-term effects of feeding a single ionophore (monensin) or rotation of 2 ionophores (monensin and lasalocid) on enteric CH4 emissions were evaluated in 36 Angus yearling steers (328 +/- 24.9 kg of BW) over a 16-wk period. Steers were randomly assigned to 6 dietary treatments of 6 steers each. The 6 diets were low-concentrate without ionophore supplementation, low-concentrate with monensin supplementation, low-concentrate with a 2-wk rotation of monensin and lasalocid supplementation, high-concentrate without ionophore supplementation, high-concentrate with monensin supplementation, and high-concentrate with a 2-wk rotation of monensin and lasalocid supplementation. Daily enteric CH4 emissions, as measured using the SF(6) tracer gas technique, ranged from 54.7 to 369.3 L/steer daily. Supplementing ionophores decreased (P < 0.05) enteric CH4 emissions, expressed as liters per kilogram of DMI or percentage of GE intake, by 30% for the first 2 wk and by 27% for the first 4 wk, for cattle receiving the high-concentrate and low-concentrate diets, respectively. Cattle fed a rotation of ionophores did not (P > 0.05) exhibit a greater decrease and did not (P > 0.05) have a longer period of depressed enteric CH4 emissions compared with cattle receiving monensin only. Ionophore supplementation did not (P > 0.05) alter total ruminal fluid VFA concentration; however, the acetate:propionate ratio and ammonia-N concentration in ruminal fluid were decreased (P < 0.001) from the time that ionophores were introduced to the time they were removed from the diets. Both monensin and the rotation of monensin and lasalocid decreased (P < 0.001) total ciliate protozoal populations by 82.5% in the first 2 wk and by 76.8% in the first 4 wk during which they were supplemented in the high-concentrate and low-concentrate diets, respectively. Eight multiparous lactating Holstein cows (initial milk yield 23.4 +/- 2.2 kg/d) were assigned to 4 dietary treatments in a replicated 4 x 4 Latin square design: a control diet (C) consisting of corn silage (59%), grass hay (6%), and concentrate (35%) and the same diet with crude linseed (CLS), extruded linseed (ELS), or linseed oil (LSO) at the same FA level (5.7% of dietary DM). Each experimental period lasted 4 wk. All the forms of linseed FA significantly decreased daily CH(4) emissions (P < 0.001) but to different extents (-12% with CLS, -38% with ELS, -64% with LSO) compared with C. The same ranking among diets was observed for CH(4) output expressed as a percentage of energy intake (P < 0.001) or in grams per kilogram of OM intake (P < 0.001). Methane production per unit of digested NDF was similar for C, CLS, and ELS but was less for LSO (138 vs. 68 g/kg of digested NDF, respectively; P < 0.001). Measured as grams per kilogram of milk or fat-corrected milk yield, methane emission was similar for C and CLS and was less for ELS and LSO (P < 0.001), LSO being less than ELS (P < 0.01). Total tract NDF digestibility was significantly less (P < 0.001) for the 3 supplemented diets than for C (-6.8% on average; P < 0.001). Starch digestibility was similar for all diets (mean 93.5%). Compared with C, DMI was not modified with CLS (P > 0.05) but was decreased with ELS and LSO (-3.1 and -5.1 kg/d, respectively; P < 0.001). Milk yield and milk fat content were similar for LSO and ELS but less than for C and CLS (19.9 vs. 22.3 kg/d and 33.8 vs. 43.2 g/kg, on average, respectively; P < 0.01 and P < 0.001). Linseed FA offer a promising dietary means to depress ruminal methanogenesis. The form of presentation of linseed FA greatly influences methane output from dairy cows. The negative effects of linseed on milk production will need to be overcome if it is to be considered as a methane mitigation agent. Six spayed Angus heifers (238 +/- 13.3 kg of initial BW) and 6 Angus steers (207 +/- 8.2 kg of initial BW) were each assigned to 2 squares. The measured condensed tannin content of the extract was 91%, and the basal diet contained 70% forage (DM basis). Feeding quebracho tannin extract had no effect on BW, ADG, or nutrient intakes. Furthermore, it had no effect on DM, energy, or fiber (ADF and NDF) digestibility, but apparent digestibility of CP decreased linearly (P < 0.001) by 5 and 15% with 1 and 2% quebracho tannin extract, respectively. Two Latin square experiments were conducted. In Exp. 1, six beef heifers (19 +/- 1 mo old) were assigned in a duplicated Latin square to 1 of 3 dietary concentrations of MA on a DMI basis (0%, MA-0; 3.75%, MA-3.75; or 7.5%, MA-7.5) over 3 periods. In Exp. 2, four rumen-fistulated steers (48 +/- 1 mo old) were assigned to 1 of 4 dietary concentrations of MA (0%, MA-0; 2.5%, MA-2.5; 5.0%, MA-5.0; or 7.5%, MA-7.5) on a DMI basis, over 4 periods. Both experimental diets consisted of grass silage and pelleted concentrate (containing MA). Silage was fed ad libitum once daily (a.m.), whereas concentrate was fed twice daily (a.m. and p.m.) with the aim of achieving a total DMI of 40:60 silage:concentrate. In both Exp. 1 and 2, experimental periods consisted of 28 d, incorporating a 13-d acclimatization, a 5-d measurement period, and a 10-d washout period. In Exp. 1, enteric CH(4), feed apparent digestibility, and feed intake were measured over the 5-d measurement period. In Exp. 2, rumen fluid was collected on d 16 to 18, immediately before (a.m.) feeding and 2, 4, 6, and 8 h thereafter. Rumen pH was determined and samples were taken for protozoa count, VFA, and ammonia analysis. Enteric CH(4) emissions were estimated by using the sulfur hexafluoride tracer technique and feed apparent digestibility was estimated by using chromic oxide as an external marker for fecal output. In a grazing experiment, 60 Holstein-Friesian cows were assigned randomly to 1 of 2 groups (control or monensin). Cows in the monensin group received 2 controlled-release capsules, with the second capsule administered 130 d after the first. Milk production was measured for 100 d following insertion of each capsule. The sulfur hexafluoride tracer gas technique was used to measure enteric methane emissions for 4 d starting on d 25 and 81 after insertion of the first capsule, and on d 83 after insertion of the second capsule. All cows grazed together as a single herd on a predominantly ryegrass sward and received 5 kg/d of grain (as-fed basis). In a second experiment, 7 pairs of lactating dairy cows (control and monensin) were used to determine the effects of monensin controlled-release capsules on methane emissions and dry matter intake. Methane emissions were measured on d 75 after capsule insertion by placing cows in respiration chambers for 3 d. Cows received fresh ryegrass pasture harvested daily and 5 kg/d of grain. The release rate of monensin from the capsules used in both experiments was 240 +/- 0.072 mg/d, determined over a 100-d period in ruminally cannulated cows. The monensin dose was calculated to be 12 to 14.5 mg/kg of dry matter intake. There was no effect of monensin on methane production in either the grazing experiment (g/d, g/kg of milk solids) or the chamber experiment (g/d, g/kg of dry matter intake). In the grazing study, there was no effect of monensin on milk yield, but monensin increased milk fat yield by 51.5 g/d and tended to increase milk protein yield by 18.5 g/d. An additional objective was to evaluate effects on milk production and rumen function, because a commensurate improvement in milk production could lead to adoption of monensin as a profitable strategy for methane abatement. Two experiments were conducted (grazing and respiratory chambers) and in both experiments monensin (471 mg/d) was topdressed on 4 kg (dry matter)/d of rolled barley grain offered in a feed trough twice daily at milking times. In the grazing experiment, 50 Holstein-Friesian cows were assigned randomly to 1 of 2 groups (control or monensin). Cows grazed together as a single herd on a predominantly ryegrass sward and received monensin over a 12-wk period, during which time measurements of milk production and body weight change were made. The SF(6) tracer technique was used to estimate methane production of 30 of the 50 cows (15 control cows and 15 monensin cows) for 3 consecutive days in wk 3, 5, 8, and 12 of treatment. Samples of rumen fluid were collected per fistula from 8 of the 50 cows (4 per diet) on 2 consecutive days in wk 3, 5, 8, and 12 of treatment and analyzed for volatile fatty acids and ammonia-N. In the metabolic chamber experiment, 10 pairs of lactating dairy cows (control and monensin) were used to determine the effects of monensin on methane emissions, dry matter intake, milk production, and body weight change over a 10-wk period. Identical diets were fed in both experiments. The mixture of feed additives investigated contained lauric acid, myristic acid, linseed oil, and calcium fumarate. These additives were included at 0.4, 1.2, 1.5, and 0.7% of dietary dry matter, respectively (treatment ADD). Experimental fat sources were exchanged for a rumen inert source of fat in the control diet (treatment CON) to maintain isolipidic rations. Cows (experiment 1, n=20; experiment 2, n=12) were fed restricted amounts of feed to avoid confounding effects of dry matter intake on methane production. In experiment 1, methane production and energy balance were studied using open-circuit indirect calorimetry. In experiment 2, 10 rumen-fistulated animals were used to measure rumen fermentation characteristics. In both experiments animal performance was monitored. The inclusion of dietary additives decreased methane emissions (g/d) by 10%. Milk yield and milk fat content tended to be lower for ADD in experiment 1. Twenty-four lactating Holstein dairy cows (1.46 +/- 0.17 parity; 620 +/- 5.9 kg of live weight; 92.5 +/- 2.62 d in milk) housed in a tie-stall facility were used in the study. The study was conducted as paired comparisons in a completely randomized design with repeated measurements in a color-coded, double-blind experiment. The cows were paired by parity and days in milk and allocated to 1 of 2 treatments: 1) the regular milking cow total mixed ration (TMR) with a forage-to-concentrate ratio of 60:40 (control TMR; placebo premix) vs. a medicated TMR (monensin TMR; regular TMR + 24 mg of Rumensin Premix/kg of dry matter) fed ad libitum. The animals were fed and milked twice daily (feeding at 0830 and 1300 h; milking at 0500 and 1500 h) and CH4 production was measured prior to introducing the treatments and monthly thereafter for 6 mo using an open-circuit indirect calorimetry system. Monensin reduced CH4 production by 7% (expressed as grams per day) and by 9% (expressed as grams per kilogram of body weight), which were sustained for 6 mo (mean, 458.7 vs. 428.7 +/- 7.75 g/d and 0.738 vs. 0.675 +/- 0.0141, control vs. monensin, respectively). Monensin reduced milk fat percentage by 9% (3.90 vs. 3.53 +/- 0.098%, control vs. monensin, respectively) and reduced milk protein by 4% (3.37 vs. 3.23 +/- 0.031%, control vs. monensin, respectively). Monensin did not affect the dry matter intake or milk yield of the cows. Eight ruminally cannulated Holstein cows were allocated to 2 dietary treatments in a crossover design. Treatments were control (no yeast culture) and XP (yeast culture, fed at 56 g/head per day; XP, Diamond V Mills Inc., Cedar Rapids, IA). Dry matter intake, milk yield, milk composition, and body weight were similar between treatments. Milk urea nitrogen concentration was also not affected by treatment. Rumen pH was similar between the control and XP treatments, but rumen ammonia concentration tended to be lower with XP than with the control. Treatment had no effect on concentrations of total or individual volatile fatty acids, protozoal counts, polysaccharide-degrading activities (except amylase activity that tended to be increased by XP), or methane production in the rumen. Urinary N losses did not differ significantly between treatments, but allantoin and total purine derivative excretions and the estimated microbial N outflow from the rumen tended to be increased by XP compared with the control treatment. Total-tract apparent digestibility of dietary nutrients was not affected by XP. Milk fatty acid composition was also not altered by XP supplementation. Lambs were gradually introduced to nitrate and sulfate in a corn silage-based diet over a period of 4 wk, and methane production was subsequently determined in respiration chambers. Diets were given at 95% of the lowest ad libitum intake observed within one block in the week before methane yield was measured to ensure equal feed intake of animals between treatments. All diets were formulated to be isonitrogenous. Methane production decreased with both supplements (nitrate: -32%, sulfate: -16%, and nitrate+sulfate: -47% relative to control). The decrease in methane production due to nitrate feeding was most pronounced in the period immediately after feeding, whereas the decrease in methane yield due to sulfate feeding was observed during the entire day. Methane-suppressing effects of nitrate and sulfate were independent and additive. The highest methemoglobin value observed in the blood of the nitrate-fed animals was 7% of hemoglobin. When nitrate was fed in combination with sulfate, methemoglobin remained below the detection limit of 2% of hemoglobin. Dietary nitrate decreased heat production (-7%), whereas supplementation with sulfate increased heat production (+3%). Feeding nitrate or sulfate had no effects on volatile fatty acid concentrations in rumen fluid samples taken 24h after feeding, except for the molar proportion of branched-chain volatile fatty acids, which was higher when sulfate was fed and lower when nitrate was fed, but not different when both products were included in the diet. The total number of rumen bacteria increased as a result of sulfate inclusion in the diet. The number of methanogens was reduced when nitrate was fed. Enhanced levels of sulfate in the diet increased the number of sulfate-reducing bacteria. The number of protozoa was not affected by nitrate or sulfate addition. In experiment 1, a control diet (CON1) and diets supplemented with 56 mg of DADS/kg of dry matter (DM), 3g of YP/kg of DM, or 25 g of CAFU/kg of DM were evaluated. In experiment 2, an inert saturated fat source in the control diet (CON2) was exchanged isolipidically for an extruded linseed source (100g/kg of DM; UNSAT) or a mixture of C8:0 and C10:0 (MCFA; 20.3g/kg of DM). In experiment 2, a higher inclusion level of DADS (200mg/kg of DM) was also tested. Both experiments were conducted using 40 lactating Holstein-Friesian dairy cows. Cows were adapted to the diet for 12 d and were subsequently kept in respiration chambers for 5 d to evaluate methane production, diet digestibility, energy balance, and animal performance. Feed intake was restricted to avoid confounding effects of possible differences in ad libitum feed intake on methane production. Feed intake was, on average, 17.5 and 16.6 kg of DM/d in experiments 1 and 2, respectively. None of the additives reduced methane production in vivo. Methane production in experiment 1 was 450, 453, 446, and 423 g/d for CON1 and the diets supplemented with DADS, YP, and CAFU, respectively. In experiment 2, methane production was 371, 394, 388, and 386 g/d for CON2 and the diets supplemented with UNSAT, MCFA, and DADS, respectively. No effects of the additives on energy balance or neutral detergent fiber digestibility were observed. The addition of MCFA increased milk fat content (5.38% vs. 4.82% for control) and fat digestibility (78.5% vs. 59.8% for control), but did not affect milk yield or other milk components.
