In ARD-affected orchards, uneven growth is usually attributed to the patchy distribution of the ARD-causing agent. But, variability in soil properties can also cause differing growth, either directly or by influencing biota involved in ARD. Evaluating phenomena under field conditions that vary at a small scale (such as ARD) is a challenging task. Here, we aimed to identify the drivers of small-scale growth variations.  <br/>\n\n<b>Methods:</b> We used a pair-wise design where neighboring trees within two commercial orchards were chosen based on maximum difference in stem diameter growth. With this spatially explicit approach, large-scale site heterogeneity as a reason for differing growth was minimized. Pairs in the plots with grass and Tagetes patula as pre-culture were chosen. <br/>\n\n<b>Results:</b> At one site, neighbors with differing growth were separated due to high particulate organic matter content, likely resulting from localized tree shredding. The worse-growing partners exhibited higher phytoalexin levels, which are general indicators of biotic stress. However, its cause remained unidentified. Amplicon sequencing was performed to assess rhizosphere bacterial community composition (data in the NCBI Sequence Read Archive (SRA) under Bioproject PRJNA1295795). Although microbiome differences were the most likely explanation for the stress response, no compositional changes were detected. At the second site, no differences depending on tree growth occurred.  <br/>\n\n<b>Conclusion:</b> Overall, our work demonstrated that sampling neighboring trees with varying stem growth can identify co-occurring differences in related variables. It was established in autumn 1998 at the experimental station of the University of Rostock. The experimental station is located in Northern Germany in a maritime-influenced area about 15 km south of the Baltic Sea shore (54\u00b03\u203241.47\u2033N; 12\u00b05\u20325.59\u2033E). The average annual temperature is 8.1 \u00b0C and the mean annual precipitation is about 600 mm. The soil texture is loamy sand containing 1.1% Corg and the soil type is a Stagnic Cambisol according to the World Reference Base for Soil Resources. The initial double-lactate-soluble P content (DL-P) of 42.2 mg P kg\u22121 soil indicated a suboptimal P supply according to the soil P classification of the German federal state of\nMV. The fields investigated were Wanna Sand (WS), Wanna Sand without biogas slurry (WSo), Rostock Sand (RS) and Wanna Marsch (WM). On the WS and WM fields, 30 m\u00b3 of biogas slurry was applied before the experiment. Starter fertilization was carried out with diammonium phosphate (DAP), microgranular fertilizer (MG) and without (control). In addition, a microorganism preparation (MO) consisting of three mycorrhizal species: R. irregulare, F. mosseae, F. caledonium and Bacillus atrophaeus Abi05 was applied to half of the plots. Soil samples were taken between April and August each year. During the vegetation period, the soil was sampled in and between the maize rows. At some sampling times, additional mixed samples were taken plot by plot. Before the experimental setup samples were taken in blocks or randomly. The abundances of the following microorganism groups were determined using qPCR: Acidobacteria, Actinobacteria, Alpha-Proteobacteria, Beta-Proteobacteria, Cyanobacteria, Firmicutes, Alternaria, Fusarium, Bacillus atrophaeus Abi05 and total fungal population. In addition to the soil samples, root samples were also taken at certain times. The rate of mycorrhization was determined and in some cases DNA was extracted and qPCR measurements were carried out.
