Increased atmospheric CO2 usually enhances photosynthetic ability and growth of trees. To understand how increased CO2 affects below-ground part of trees under varied soil condition, we investigated the responses of the fine root (diameter <2\u00a0mm) dynamics of Japanese white birch (Betula platyphylla var. japonica) which was planted in 2010. The three-year-old birch seedlings were grown in four experimental treatments comprising two levels of CO2, i.e., ambient: 380\u2013390 and elevated: 500\u00a0\u03bcmol\u00a0mol\u22121, in combination with two kinds of soil: brown forest (BF) soil and volcanic ash (VA) soil which has few nutrients. The growth and turnover of fine roots were measured for 3\u00a0years (2011\u20132013) using the Mini-rhizotron. In the first observation year, live fine root length (standing crop) in BF soil was not affected by CO2 treatment, but it was reduced by the elevated CO2 from the second observation year. In VA soil, live fine root length was reduced by elevated CO2 for all 3\u00a0years. Fine root turnover tended to decrease under elevated CO2 compared with ambient in both soil types during the first and second observation years. Turnover of fine root production and mortality was also affected by the two factors, elevated CO2 and different soil types. Median longevity of fine root increased under elevated CO2, especially in VA soil at the beginning, and a shorter fine root lifespan appeared after 2\u00a0years of observation (2011\u20132012). These results suggest that elevated CO2 does not consistently stimulate fine root turnover, particularly during the plant seedlings stage, as it may depend on the costs and benefits of constructing and retaining roots. Calcium-carbonate additions increased soil respiration in the field plots in the SHF; in laboratory incubations, soil respiration was higher in the SHF when soils were fertilised with N, and in THF and LERF after S additions. The addition of N alone or in different combinations generally induced a net immobilisation of soil N. Net nitrification increased during the incubation in SHF and THF soils fertilised with N+P, and in LERF soils fertilised with either N, or P, or CaCO3. In a field experiment using ingrowth bags, a higher fine root production was observed in all forest types when bags were fertilised with CaCl2 or CaCO3, suggesting that Ca may be a limiting nutrient in these soils. Calcium-carbonate addition in a glasshouse bioassay experiment with rice showed an overall positive effect on the survival and growth of the seedlings. In other treatments where soil pH was not raised, the rice showed acute toxicity symptoms, poor root and shoot growth and high mortality. Similar results were yielded in a field experiment, using naturally established seedlings in the field plots in SHF, THF and LERF. The principal habitat for microorganisms is within rocks (endolithic). In this environment, microorganisms are provided with protection against sub-zero temperatures, rapid thermal fluctuations, extreme dryness, and ultraviolet and solar radiation. Endolithic communities include lichen, algae, fungi, and a diverse array of bacteria.               Chloroflexota               is among the most abundant bacterial phyla present in these communities. Among the               Chloroflexota               are four novel classes of bacteria, here named               Candidatus               Spiritibacteria class. nov. (=UBA5177),               Candidatus               Martimicrobia class. nov. (=UBA4733),               Candidatus               Tarhunnaeia class. nov. (=UBA6077), and               Candidatus               Uliximicrobia class. nov. (=UBA2235). We retrieved 17 high-quality metagenome-assembled genomes (MAGs) that represent these four classes. Based on genome predictions, all these bacteria are inferred to be aerobic heterotrophs that encode enzymes for the catabolism of diverse sugars. These and other organic substrates are likely derived from lichen, algae, and fungi, as metabolites (including photosynthate), cell wall components, and extracellular matrix components. The majority of MAGs encode the capacity for trace gas oxidation using high-affinity uptake hydrogenases, which could provide energy and metabolic water required for survival and persistence. Furthermore, some MAGs encode the capacity to couple the energy generated from H               2               and CO oxidation to support carbon fixation (atmospheric chemosynthesis). All encode mechanisms for the detoxification and efflux of heavy metals. Certain MAGs encode features that indicate possible interactions with other organisms, such as Tc-type toxin complexes, hemolysins, and macroglobulins.             </p>                            IMPORTANCE               <p>                 The ice-free McMurdo Dry Valleys of Antarctica are the coldest and most hyperarid desert on Earth. It is, therefore, the closest analog to the surface of the planet Mars. Bacteria and other microorganisms survive by inhabiting airspaces within rocks (endolithic). We identify four novel classes of phylum                 Chloroflexota                 , and, based on interrogation of 17 metagenome-assembled genomes, we predict specific metabolic and physiological adaptations that facilitate the survival of these bacteria in this harsh environment\uffe2\uff80\uff94including oxidation of trace gases and the utilization of nutrients (including sugars) derived from lichen, algae, and fungi. Among environmental pollutants, polystyrene (PS) stands out as a primary constituent of environmental plastic waste, given its widespread use and high production rates owing to its durability and user-friendly properties. The detection of polystyrene microparticles (PS-MPs) in various living organisms has been well-documented, posing a serious threat due to their potential passage into the human ecosystem. In this manuscript, we aimed to study the toxicological effects of low concentrations of pristine and photoaged PS-MPs in a murine macrophage cell line. To this purpose, PS-MPs were photoaged by indoor exposure to visible light to simulate environmental weathering due to solar irradiation (PS-MPs3h). Physical characterization revealed that the irradiation treatment results in particle degradation and the possible release of nanoparticles. Monocultures of the RAW264.7\u00a0cell line were then exposed to PS-MPs and PS-MPs3h at concentrations comparable to experimental measurements from biological samples, to assess cytotoxicity, intracellular oxidative stress, primary genotoxicity, and inflammatory effects. Significant toxicity-related outcomes were observed in cells treated with both pristine PS-MPs and PS-MPs3h even at low concentrations (0,10\u00a0\u03bcg/ml and 1\u00a0\u03bcg/ml). PS-MPs3h exhibited greater adverse effects compared to PS-MPs, including reduced cell viability, increased ROS production, elevated DNA damage, and upregulation of IL-6 and NOS2 gene expression. Don)), western hemlock (Tsuga heterophylla ([Raf.] Sarg.)) and amabilis fir (Abies amabilis ([Dougl. ex Loud] Dougl. ex Forbes)). Seedlings were planted at two different sites (forest interior: 4.5% mean above canopy photosynthetically active radiation [ACPAR], and forest edge: 41.5% mean ACPAR), in a 90-year-old, windthrow origin, unmanaged mesic western hemlock-amabilis fir stand. Seedlings were planted in 1997, and re-measured in 1998 and 2001 (after five growing seasons). To assess the effects of deer browsing on redcedar survival and growth, additional seedlings of this species were planted and protected with Vexar\u00a9 tubes. To examine for nutrient-light interactions, half of these seedlings were fertilized with N-P-K and micronutrients at planting. Western redcedar had high levels of survival after 4 years (98% in edge plots and 93% in interior plots). Redcedar seedlings in edge plots were more vigorous but were browsed more heavily than in the interior plots. At edge sites, the negative effects of the Vexar\u00a9 tubes may have been lower than their positive effects. Hemlock survival was about 50% in the stand interior but 80% in the edge plots. Amabilis fir in the interior plots had the lowest survival of the three species, with only 40% of initial seedlings surviving over the next four years, but had high survival in edge plots (95%). Height, biomass, and root collar diameter growth were significantly higher in edge plots for fir and hemlock. However, for redcedar, only biomass was significantly higher and no differences were detected for height and diameter. A common, yet untested, assumption in forecasting approaches is that species will shift beyond current range edges into new habitats as they become macroclimatically suitable, even though microhabitat variability could have overriding effects on local population dynamics. We aim to better understand the role of microhabitat in range shifts in plants through its impacts on establishment by Q1) examining microhabitat variability along large macroclimatic (i.e., elevational) gradients, Q2) testing which of these microhabitat variables explain plant recruitment and seedling survival, and Q3) predicting microhabitat suitability beyond species range limits. We transplanted seeds of 25 common tree, shrub, forb, and graminoid species across and beyond their current elevational ranges in the Washington Cascade Range, USA, along a large elevational gradient spanning a broad range of macroclimates. Over five years, we recorded recruitment, survival, and microhabitat (i.e., high resolution soil, air, and light) characteristics rarely measured in biogeographic studies. We asked whether microhabitat variables correlate with elevation, which variables drive species establishment, and whether microhabitat variables important for establishment are already suitable beyond leading range limits. We found that only 30% of microhabitat parameters covaried with elevation. We further observed extremely low recruitment and moderate seedling survival, and these were generally only weakly explained by microhabitat. Moreover, species and life stages responded in contrasting ways to soil biota, soil moisture, temperature, and snow duration. Microhabitat suitability predictions suggest that distribution shifts are likely to be species-specific, as different species have different suitability and availability of microhabitat beyond their present ranges, thus calling into question low-resolution macroclimatic projections that will miss such complexities. Seed germination of all species increased with experimental warming, suggesting that the present treeline may in part result from unsuccessful recruitment under cold conditions.</p><p> 2 Growth, biomass and survivorship of seedlings of treeline species transplanted into tundra were largely unaffected by experimental warming. However, transplanted seedlings of three species (Betula papyrifera, Picea glauca and Populus tremuloides) grew more when below\uffe2\uff80\uff90ground competition with the extant community was reduced. All three measures of transplant performance were greater in shrub tundra than in the less productive tussock or heath tundra. Establishment of trees in tundra may thus be prevented by low resource availability and competition.</p><p> 3 Two species (Alnus crispa and Populus balsamifera) had low seed germination and survivorship of germinated seeds; transplants of these species did not respond to the manipulations and lost biomass following transplanting into tundra. Isolated populations of these two species north of the present treeline in arctic Alaska probably became established during mid\uffe2\uff80\uff90Holocene warming rather than in recent times.</p><p> 4 Of all the species studied here, Picea glauca was the most likely to invade intact upland tundra. Its seeds had the highest germination rates and it was the only species whose seedlings survived subsequently. We examined individual nanoparticle-treated cells by Raman spectroscopy and analyzed Raman spectra using non-negative principal component analysis and k-means clustering. In this way, we determined dose-dependent differences between treated cells, which were reflected through the intensity change of lipid, phospholipid and RNA-related Raman modes. Performing standard biological tests for cell growth, viability and induction of apoptosis in parallel, these changes were correlated with nanoparticle-induced apoptotic processes. The cells with specific spectral characteristics, referring to non-apoptotic, but possibly autophagic cell death modality, were also detected. Additionally, Raman imaging combined with principal component and vertex component analysis was used to map the spatial distribution of biological molecules in treated and untreated cells. This work provided the description of different resulting states of the treated cells depending on the dextran-coated CeO<sub>2</sub> nanoparticles dose, which can be later used in the design of the nanoparticles for industrial or medical applications. The principal habitat for microorganisms is within rocks (endolithic). In this environment, microorganisms are provided with protection against sub-zero temperatures, rapid thermal fluctuations, extreme dryness, and ultraviolet and solar radiation. Endolithic communities include lichen, algae, fungi, and a diverse array of bacteria.               Chloroflexota               is among the most abundant bacterial phyla present in these communities. Among the               Chloroflexota               are four novel classes of bacteria, here named               Candidatus               Spiritibacteria class. nov. (=UBA5177),               Candidatus               Martimicrobia class. nov. (=UBA4733),               Candidatus               Tarhunnaeia class. nov. (=UBA6077), and               Candidatus               Uliximicrobia class. nov. (=UBA2235). We retrieved 17 high-quality metagenome-assembled genomes (MAGs) that represent these four classes. Based on genome predictions, all these bacteria are inferred to be aerobic heterotrophs that encode enzymes for the catabolism of diverse sugars. These and other organic substrates are likely derived from lichen, algae, and fungi, as metabolites (including photosynthate), cell wall components, and extracellular matrix components. The majority of MAGs encode the capacity for trace gas oxidation using high-affinity uptake hydrogenases, which could provide energy and metabolic water required for survival and persistence. Furthermore, some MAGs encode the capacity to couple the energy generated from H               2               and CO oxidation to support carbon fixation (atmospheric chemosynthesis). All encode mechanisms for the detoxification and efflux of heavy metals. Certain MAGs encode features that indicate possible interactions with other organisms, such as Tc-type toxin complexes, hemolysins, and macroglobulins.             </p>                            IMPORTANCE               <p>                 The ice-free McMurdo Dry Valleys of Antarctica are the coldest and most hyperarid desert on Earth. It is, therefore, the closest analog to the surface of the planet Mars. Bacteria and other microorganisms survive by inhabiting airspaces within rocks (endolithic). We identify four novel classes of phylum                 Chloroflexota                 , and, based on interrogation of 17 metagenome-assembled genomes, we predict specific metabolic and physiological adaptations that facilitate the survival of these bacteria in this harsh environment\uffe2\uff80\uff94including oxidation of trace gases and the utilization of nutrients (including sugars) derived from lichen, algae, and fungi. Human cancer (U-87) and normometabolic (MRC-5) cells were treated with different concentrations of larval hemolymph proteins, and the effects on cell viability were assayed 24, 48, and 72 h after treatments. Z. morio hemolymph was shown to be richer in total proteins, showing a higher antioxidant capacity and lipid peroxidation level than T. molitor hemolymph, which was richer in total carbohydrates. Cytotoxicity assays showed that T. molitor and Z. morio hemolymphs differently affect the viability of U-87 and MRC-5 cells in cell type-, dose-, and time-dependent manners. In this study, selected biochemical properties and cell toxicity of larval hemolymph from two mealworm species, Tenebrio molitor and Zophobas morio, were analyzed. Total proteins and carbohydrates, antioxidant capacity, and the level of lipid peroxidation were determined. Human cancer (U-87) and normometabolic (MRC-5) cells were treated with different concentrations of larval hemolymph proteins, and the effects on cell viability were assayed 24, 48, and 72 h after treatments. Z. morio hemolymph was shown to be richer in total proteins, showing a higher antioxidant capacity and lipid peroxidation level than T. molitor hemolymph, which was richer in total carbohydrates. Cytotoxicity assays showed that T. molitor and Z. morio hemolymphs differently affect the viability of U-87 and MRC-5 cells in cell type-, dose-, and time-dependent manners. Hemolymph from both species was more cytotoxic to U-87 cells than to MRC-5 cells, which was particularly prominent after 48 h.
