However, the precise relationships between soil properties and microbial diversity are poorly known. Here, we study the effect of agricultural management on soil microbial diversity and C turnover in tropical grassland of north-eastern Laos. Three years after native grassland conversion into agricultural land, we compared soils from five land use management systems: one till versus two no-till rotational cropping systems, one no-till improved pasture and the natural grassland. Soils were incubated in microcosms during 64 days at optimum temperature and humidity. Bacterial and fungal diversity were evaluated by metagenomic 454-pyrosequencing of 16S and 18SrRNA genes, respectively. Changes in soil respiration patterns were evaluated by monitoring 12C- and 13C-CO2 release after soil amendment with 13C-labelled wheat residues. Results show that residue mineralization increased with bacterial richness and diversity in the tilled treatment 7 days after soil amendment. Native soil organic C mineralization and priming effect increased with fungal richness and diversity in improved pasture and natural grassland. No-till cropping systems represented intermediate situations between tillage and pasture systems. Our findings evidence the potential of controlling soil microbial diversity by agricultural practices to improve soil biological properties. This work reviewed seven important European LCA studies in a sensitivity analysis approach in order to get a better understanding of the roots of such a debate for three major biofuels in European production: rape methyl ester and ethanol from wheat and sugar beet. Global trends and variability of energy and GHG balances were depicted and completed with a sensitivity analysis carried out for each methodological and data parameter, which allowed making recommendations on the carrying out of LCA in a policy-making or a biofuels comparison context. Methodological choices, and especially allocation rule, appeared as key elements for results variation with influences on balances up to 149%; system expansion approach was identified as the most relevant rule since it integrates the market potential and the environmental interest of by-products promotion, which was pointed out as a crucial point for biofuels sustainability. The influence of local specificity for cultivation data was evaluated up to 167%, which puts too large geographical coverage in question. Modelling uncertainties due to N2O emissions from soils showed influences from 17 to 46%, which represents a crucial challenge for research and for LCA results accuracy. Approximations evaluation pointed out the need to integrate agricultural machinery into the assessment. Results showed that application of sufficient slaked lime to initially increase the pH of the topsoil by 1 unit caused an increase in pH to 5 cm deeper than the layer of application as a result of bicarbonate leaching. With leaching of Ca from slaked lime or gypsum from the topsoil to the subsoil there was a decrease in exchangeable Al in the subsoil. Surface application of slaked lime or gypsum or both decreased the activity of toxic Al and increased AlSO4+ activity in the subsoil solution. The Ca added in slaked lime or gypsum was accounted for by the increase in exchangeable Ca over the soil profile and the leaching loss. By contrast there was a negative balance of extractable sulfate and aluminum in the soil, indicating the formation of precipitates. There was little mineralisation of N and formation of NO3- under the conditions of the experiment.
