The process was investigated at bench scale under different oxidant doses (0-50\u00a0mg\u00a0L-1) with regard to its capacity to degrade a mixture of antibiotics (i.e. ampicillin, clarithromycin, erythromycin, ofloxacin, sulfamethoxazole, tetracycline and trimethoprim) with an initial individual concentration of 100\u00a0\u03bcg\u00a0L-1. The process was optimized with respect to the oxidant dose. Under the optimum conditions, the inactivation of selected bacteria and antibiotic resistant bacteria (ARB) (i.e. faecal coliforms, Enterococcus spp., Pseudomonasaeruginosa and total heterotrophs), and the reduction of the abundance of selected antibiotic resistance genes (ARGs) (e.g. blaOXA, qnrS, sul1, tetM) were investigated. Also, phytotoxicity against three plant species, ecotoxicity against Daphnia magna, genotoxicity, oxidative stress and cytotoxicity were assessed. Apart from chemical actinometry, computational fluid dynamics (CFD) modelling was applied to estimate the fluence rate. For the given wastewater quality and photoreactor type used, 40\u00a0mg\u00a0L-1 H2O2 were required for the complete degradation of the studied antibiotics after 18.9\u00a0J\u00a0cm-2. Total bacteria and ARB inactivation was observed at UV doses <1.5\u00a0J\u00a0cm-2 with no bacterial regrowth being observed after 24\u00a0h. The abundance of most ARGs was reduced at 16\u00a0J\u00a0cm-2. The process produced a final effluent with lower phytotoxicity compared to the untreated wastewater. The toxicity against Daphnia magna was shown to increase during the chemical oxidation. Although genotoxicity and oxidative stress fluctuated during the treatment, the latter led to the removal of these effects. Quantitative PCR (qPCR) is a standard approach to detect and quantify ARGs in environmental compartments. However, the comparison of gene quantification reported by different laboratories is challenging since data are predominantly obtained under non- harmonized conditions, using different qPCR protocols. Objectives: The aim of this study was to develop and calibrate standardized qPCR procedures for quantification of key ARGs, analyzing the same samples with common protocols and distinct equipment, reagents batches and operators. Methods: Treated wastewater from three European countries were processed immediately after collection and transported to the laboratory for total DNA extraction. DNA extracts from each sample were pooled and aliquots were distributed by five partners involved in the calibration procedure. The genes 16S rRNA, vanA, blaTEM, qnrS, sul1, blaCTXM-32 and intI1 were analyzed using harmonized qPCR protocols and the constructed pNORM1 plasmid, which contains fragments of the seven targeted genes, was used for generating standard curves. Conclusions: The 16S rRNA gene was the most abundant, followed by sul1, intI1, qnrS and blaTEM. Quantifications made by different partners were reproducible and inter-laboratory variation was &lt; 20%. The notorious exception was for the qnrS gene, and therefore protocol improvement is recommended. The genes blaCTXM-32 and vanA were below the limit of quantification in most or all of the samples analyzed. The biochars used were obtained from two contrasting feedstocks, namely wood chips and wheat straw, by gasification under high temperature conditions, and were applied in a greenhouse experiment either as pure or as nutrient-activated biochar obtained by incubation with digestate. The results of the experiment showed that specific genotypes as well as different treatments with biochar have significant effects on plant response when looking at shoot traits related to growth. The evaluated genotypes could be clustered in two main distinct groups presenting, respectively, significantly increasing (Duilio, Iride, and Saragolla varieties) and decreasing (Marco Aurelio and Grecale varieties) values of projected shoot system area (PSSA), fresh weight (FW), dry weight (DW), and plant water loss by evapotranspiration (ET). All these traits were correlated with Pearson correlation coefficients ranging from 0.74 to 0.98. Concerning the treatment effect, a significant alteration of the mentioned plant traits was observed when applying biochar from wheat straw, characterized by very high electrical conductivity (EC), resulting in a reduction of 34.6% PSSA, 43.2% FW, 66.9% DW, and 36.0% ET, when compared to the control. Interestingly, the application of the same biochar after nutrient spiking with digestate determined about a 15\uffe2\uff80\uff9330% relief from the abovementioned reduction induced by the application of the sole pure wheat straw biochar. Understanding the underlying mechanisms is critical for sub\uffe2\uff80\uff90seasonal to annual wildfire predictions since the magnitude of certain teleconnection climate modes (TCMs) intensifies or they may undergo phase shifts. Here, we study how TCMs govern wildfire activity and compare the effects of weather and fuels in mediating the influence of TCMs on wildfires. Globally, burned area (BA) is predictable by a single TCM in 25.4% of the burnable (vegetated) regions, with Australia and eastern Siberia identified as the two hot spots with the highest probability out of a total of 10. Tropical oceans are the primary sources of teleconnection\uffe2\uff80\uff90driven variability in global BA. Our study finds that in dryland hot spots such as Australia, the Horn of Africa, and the northern Middle East, the lagged mediating effects of fuels outweigh the immediate mediating effects of weather. Whereas in hot spots with dense vegetation, like northeastern South America and Southeast Asia, the immediate mediating effects of weather are generally more dominant. In other hot spots, fuels can still serve as a key pathway through which specific TCMs influence wildfire activity. This study highlights the important role of fuels in transmitting the delayed impacts of TCMs\uffe2\uff80\uff90induced weather anomalies on regional wildfire activity. These climatic grids do not reflect conditions below vegetation canopies and near the ground surface, where critical ecosystem functions occur and most terrestrial species reside. Here, we provide global maps of soil temperature and bioclimatic variables at a 1\uffe2\uff80\uff90km2resolution for 0\uffe2\uff80\uff935 and 5\uffe2\uff80\uff9315\uffc2\uffa0cm soil depth. These maps were created by calculating the difference (i.e. offset) between in situ soil temperature measurements, based on time series from over 1200 1\uffe2\uff80\uff90km2pixels (summarized from 8519 unique temperature sensors) across all the world's major terrestrial biomes, and coarse\uffe2\uff80\uff90grained air temperature estimates from ERA5\uffe2\uff80\uff90Land (an atmospheric reanalysis by the European Centre for Medium\uffe2\uff80\uff90Range Weather Forecasts). We show that mean annual soil temperature differs markedly from the corresponding gridded air temperature, by up to 10\uffc2\uffb0C (mean\uffc2\uffa0=\uffc2\uffa03.0\uffc2\uffa0\uffc2\uffb1\uffc2\uffa02.1\uffc2\uffb0C), with substantial variation across biomes and seasons. Over the year, soils in cold and/or dry biomes are substantially warmer (+3.6\uffc2\uffa0\uffc2\uffb1\uffc2\uffa02.3\uffc2\uffb0C) than gridded air temperature, whereas soils in warm and humid environments are on average slightly cooler (\uffe2\uff88\uff920.7\uffc2\uffa0\uffc2\uffb1\uffc2\uffa02.3\uffc2\uffb0C). The observed substantial and biome\uffe2\uff80\uff90specific offsets emphasize that the projected impacts of climate and climate change on near\uffe2\uff80\uff90surface biodiversity and ecosystem functioning are inaccurately assessed when air rather than soil temperature is used, especially in cold environments. The global soil\uffe2\uff80\uff90related bioclimatic variables provided here are an important step forward for any application in ecology and related disciplines.
