In this study we investigate the influence of no-tillage and soil characteristics on the efficiency of a urease inhibitor (N-(n-butyl)thiophosphoric triamide, NBPT) and a nitrification inhibitor (diciandiamide, DCD) in decreasing ammonia volatilization from urea and ammonium nitrate (AN), respectively.The results indicate that ammonia volatilization in soils amended with urea was significantly higher than in those fertilized with AN. Likewise, the main soil factors affecting ammonia volatilization from urea are clay and sand soil contents. While clay impedes ammonia volatilization, sand favours it. The presence of organic residues on soil surface (no-tillage) tends to increase ammonia volatilization from urea, although this fact depended on soil type. The presence of NBPT in urea fertilizer significantly reduced soil ammonia volatilization. This action of NBPT was negatively affected by acid soil pH and favoured by soil clay content.The presence of organic residues on soil surface amended with urea increased ammonia volatilization, and was particularly high in sandy compared with clay soils. Application of NBPT reduced ammonia volatilization although its efficiency is reduced in acid soils. There is a growing interest in the formulations of coating chemical fertilizers with both UI and NI. However, limited information is available on the combined use of UI and NI applied with urea fertilizer. Therefore the aim of this study was to investigate the effects of treating urea with both UI and NI to minimize NH3 volatilization. Two experiments were set up in volatilization chambers under controlled conditions to examine this process. In the first experiment, UR was treated with the urease inhibitor NBPT [N-(n-butyl) thiophosphoric acid triamide] at a rate of 1060\u00a0mg\u00a0kg\u22121 urea and/or with the nitrification inhibitor DCD (dicyandiamide) at rates equivalent to 5 or 10% of the urea N. A randomized experimental design with five treatments and five replicates was used: 1) UR, 2) UR\u00a0+\u00a0NBPT, 3) UR\u00a0+\u00a0DCD 10%, 4) UR\u00a0+\u00a0NBPT\u00a0+\u00a0DCD 5%, and 5) UR\u00a0+\u00a0NBPT\u00a0+\u00a0DCD 10%. The fertilizer treatments were applied to the surface of an acidic Red Latosol soil moistened to 60% of the maximum water retention and placed inside volatilization chambers. Controls chambers were added to allow for NH3 volatilized from unfertilized soil or contained in the air that swept over the soil surface. The second experiment had an additional treatment with surface-applied DCD. The chambers were glass vessels (1.5\u00a0L) fit with air inlet and outlet tubings to allow air to pass over the soil. Ammonia volatilized was swept and carried to a flask containing a boric acid solution to trap the gas and then measured daily by titration with a standardized H2SO4 solution. Continuous measurements were recorded for 19 and 23 days for the first and second experiment, respectively. The soil samples were then analyzed for UR\u2013,       NH   4  +   \u2013    , and       NO   3  \u2212   \u2013  N    . Losses of NH3 by volatilization with unamended UR ranged from 28 to 37% of the applied N, with peak of losses observed the third day after fertilization. NBPT delayed the peak of NH3 losses due to urease inhibition and reduced NH3 volatilization between 54 and 78% when compared with untreated UR. Up to 10 days after the fertilizer application, NH3 losses had not been affected by DCD in the UR or the UR\u00a0+\u00a0NBPT treatments; thereafter, NH3 volatilization tended to decrease, but not when DCD was present. As a consequence, the addition of DCD caused a 5\u201316% increase in NH3 volatilization losses of the fertilizer N applied as UR from both the UR and the UR\u00a0+\u00a0NBPT treatments. Because the effectiveness of NBPT to inhibit soil urease activity was strong only in the first week, it could be concluded that DCD did not affect the action of NBPT but rather, enhanced volatilization losses by maintaining higher soil       NH   4  +      concentration and pH for a longer time. In this context, a field experiment was carried out with a barley crop (Hordeum vulgare L.) under Mediterranean conditions to test the effectiveness of the urease inhibitor N-(n-butyl) thiophosphoric triamide (NBPT) on reducing these gaseous N losses from surface applied urea. Crop yield, soil mineral N concentrations, dissolved organic carbon (DOC), denitrification potential, NH(3), N(2)O and NO fluxes were measured during the growing season. Application of N fertilizer slightly decreased CH4 uptake by soil. Direct N2O emissions derived from recommended urea application was 0.39% of the annual urea-N input. Both straw and manure had relatively low N2O emissions factors. Slow release urea had a relatively high emission factor. Addition of nitrification inhibitor reduced N2O emission by 55%. During two consecutive campaigns, the new nitrification inhibitor 2-(3,4-dimethyl-1H-pyrazol-1-yl) succinic acid isomeric mixture (DMPSA) applied with calcium ammonium nitrate (CAN) and the same fertilizer applied by drip-fertigation without the inhibitor, were evaluated and compared with CAN broadcast to the surface and irrigated with sprinklers. Concurrently, urea-based treatments such as urea-fertigation and the broadcast application of urea combined with sprinkler irrigation, with or without the urease inhibitor N-butyl thiophosphorictriamide (NBPT), were also assessed. Nitrous oxide (N2O) and nitric oxide (NO) fluxes, grain and biomass yield and yieldscaled N2O emissions ofthe differenttreatments were compared.Additionally, methane (CH4) and carbon dioxide (CO2) fluxes were measured. On average, fertigation treatments led to a mitigation of N2O emissions with respect to sprinkler irrigation by 80% and 78% for CAN and urea, respectively. With regards to inhibitor-based strategies, the use of DMPSA and NBPT reduced N2O losses by 58% and 51%, respectively, considering the average of both maize cropping seasons. Since no differences in grain yield were observed between fertilized treatments, DMPSA and fertigation treatments gave the lowest values of yield-scaled N2O emissions, leading to reductions of 63%, 71% and 78% for CAN with DMPSA, urea-fertigation and CAN-fertigation, respectively, with respect to conventional management strategies (surface broadcast application and sprinkler irrigation). Low NO emissions during the first campaign masked differences between treatments, whereas during the second season, NO losses significantly decreased in the following order: conventional treatments > inhibitors > fertigation. Comparing conventional management practices, CAN significantly decreased emissions of N oxides compared with urea, but this effect was only observed in the second maize cropping season. The moisture distribution pattern in drip plots (dry and wet areas) caused a reduction of CH4 sink (only in one of the two seasons) and respiration fluxes, in comparison to sprinkler. To evaluate this potential risk, we analyzed microbiomes and resistomes of soil and Lactuca sativa L. (lettuce) root samples from pots irrigated with tap water spiked with 0, 20, or 100 \u03bcg L-1 of a mixture of three antibiotics (Trimethoprim, Ofloxacin, Sulfamethoxazole). The presence of antibiotics induced changes in bacterial populations, particularly in soil, as revealed by 16S rDNA sequence analysis. Parallel shotgun sequencing identified a total of 56 different ARGs conferring resistance against 14 antibiotic families. Antibiotic -treated samples showed increased loads of ARGs implicated in mutidrug resistance or in both direct and indirect acquired resistance. These changes correlated with the prevalence of Xantomonadales species in the root microbiomes. We interpret these data as indicating different strategies of soil and root microbiomes to cope with the presence of antibiotics, and as a warning that their presence may increase the loads of ARBs and ARGs in edible plant parts, therefore constituting a potential risk for human consumers.", "keywords": ["2. In the present work, we hypothesized that the ARG load and irrigation intensity define the effect of TWW irrigation on ARG spread dynamics in soil. This hypothesis was tested using a multiphase approach: a) comparing soil from a full-scale, commercially operated, TWW irrigated field with non-irrigated soil, b) long-term sampling of the TWW irrigated field over one year with different irrigation intensities and intercepted by irrigation breaks and c) laboratory-scale soil microcosms irrigated with TWW compared to FW. Six ARGs, the integrase gene intI1 and the 16S rRNA were quantified using qPCR. In addition, effects of TWW irrigation on bacterial community composition of microcosm-samples were analysed with 16S rRNA amplicon sequencing. The genes sul1, qnrS, blaOXA-58, tet(M) and intI1 were significantly more abundant in the TWW irrigated field soil, whereas blaCTX--M-32 and blaTEM, the least abundant genes in the TWW irrigation, showed higher abundance in the non-irrigated soil. The relative abundance of sul1, qnrS, blaOXA-58, tet(M) and intI1 correlated with TWW irrigation intensity and decreased during irrigation breaks. Despite the decrease, the levels of these genes remained consistently higher than the non-irrigated soil indicating persistence upon their introduction into the soil. Microcosm experiments verified observations from the field study: TWW irrigation promoted the spread of ARGs and intI1 into soil at far elevated levels compared to FW irrigation. However, the impact of TWW irrigation on 16S rRNA absolute abundance and the soil microbial community composition was negligible. Nonetheless, wastewater-derived bacteria and associated antibiotic resistance genes (ARGs) may persist below detection levels and potentially proliferate under copiotrophic conditions. To test this hypothesis, we exposed soils from microcosm, lysimeter, and field experiments to short-term enrichment in copiotroph-stimulating media. In microcosms, enrichment stimulated growth of multidrug-resistant Escherichia coli up to 2 weeks after falling below detection limits. Lysimeter and orchard soils irrigated in-tandem with either freshwater or TW were subjected to culture-based, qPCR and shotgun metagenomic analyses prior, and subsequent, to enrichment. Although native TW- and freshwater-irrigated soil microbiomes and resistomes were similar to each other, enrichment resulted in higher abundances of cephalosporin- and carbapenem-resistant Enterobacteriaceae and in substantial differences in the composition of microbial communities and ARGs. Enrichment stimulated ARG-harboring Bacillaceae in the freshwater-irrigated soils, whereas in TWW-irrigated soils, ARG-harboring \u03b3-proteobacterial families Enterobacteriaceae and Moraxellaceae were more profuse. The fitness of wastewater ARB in the complex fecal microbiota of a healthy human was investigated in feces-based microcosm assays (FMAs). FMAs were inoculated with two wastewater isolates, Escherichia coli strain A2FCC14 (MLST ST131) and Enterococcus faecium strain H1EV10 (MLST ST78), harboring the ARGs blaTEM, blaCTX, blaOXA-A and vanA, respectively. The FMAs, incubated in the presence or absence of oxygen or in the presence or absence of the antibiotics cefotaxime or vancomycin, were monitored based on cultivation, ARGs quantification and bacterial community analysis. The fecal bacterial community was dominated by members of the phyla Firmicutes, Bacteroidetes, Actinobacteria, Proteobacteria and Verrucomicrobia. The ARGs harbored by the wastewater isolates could be quantified after one week, in FMAs incubated under both aerobic and anaerobic conditions. These observations were not significantly different in FMAs incubated anaerobically, supplemented with sub-inhibitory concentrations of cefotaxime or vancomycin. The aim of this study was to investigate the effect of urease inhibitors and biological preparations in combination with nitrogen fertilizers on the productivity of aboveground maize (Zea mays L.) biomass. The highest dry matter yield of maize (24.1 t ha\u22121) was obtained with N180 fertilizer and UI ATS. UI ATS significantly increased the dry matter yield of the aboveground maize in all nitrogen fertilization backgrounds. Biochar addition did not affect the annual N2O emissions (26\uffe2\uff80\uff9328\uffc2\uffa0kg N/ha), but reduced seasonal N2O emissions during the cold period. Biochar increased soil organic C and CO2 efflux on average by 61 and 19%, respectively. Biochar addition greatly increased C gain in the acidic soil (average 11.1\uffc2\uffa0Mg C/ha) compared with treatments without biochar addition (average \uffe2\uff88\uff922.2\uffc2\uffa0Mg C/ha). Biochar amendment did not increase yield\uffe2\uff80\uff90scaled N2O emissions after application of mineral fertilizer, but it decreased yield\uffe2\uff80\uff90scaled N2O by 15% after nitrapyrin addition. This study aimed to evaluate nitrogen losses by ammonia volatilization from stabilized, slow and controlled release urea and its absorption by maize grown under no-till in an improved Cerrado soil. Four N sources were used: conventional urea, urea + N-(n-butyl) thiophosphoric triamide (NBPT), urea + Cu and B and urea coated by sulfur + polymers. These N sources were surface applied along the rows using three N doses of 100, 150 and 200 kg ha-1. No N was added to the control. Data were collected regarding N losses by volatilization, the N contents accumulated in the stubble and grains, and the yields of the stubble and grains. Stabilized urea and slow release urea were efficient for postponing the ammonia volatilization peaks. The urease inhibitors postponed the peaks for up to two days, reducing the accumulated volatilization by 18% when compared with common urea. Polymer sulfur coated urea resulted in a 37% reduction in ammonia volatilization. Increasing the N application rate to 200 kg ha-1 resulted in 16% greater yields and 37% greater N accumulation in the plants relative to the control. However, the stabilized and slow-release urea did not improve the N accumulation or yield. The effects of the incorporation of NBPT, copper (Cu+2), boric acid (H3BO3), elemental sulphur (S\uffc2\uffba), and a clay mineral from the zeolite group in powder urea - with ten different combinations of these additives - were evaluated as to N losses by volatilization and leaching. The losses in laboratory-developed formulations were compared with those of commercial fertilizers coated with the same additives (Super N, FH Nitro Mais, and FH Nitro Gold). The evaluations were made in greenhouse conditions, using a Ultisol accommodated in PVC columns. Nitrate and ammonium leaching was evaluated in the solution percolated through the soil columns. Ammonia volatilization was measured with a semi-open static chamber. The incorporation of urease inhibitors (NBPT, H3BO3, and Cu+2) into the urea granules was efficient to reduce N volatilization. Ammonia volatilization in the laboratory-developed ureas was lower than in commercial fertilizers coated with the same additives, while ammonium sulfate losses by leaching were similar. The addition of zeolite does not reduce N volatilization. A. taxiformis contains several anti-methanogenic compounds including bromoform that inhibits directly methanogenesis. The positive and adverse effects of A. taxiformis on the rumen microbiota are dose-dependent and operate in a dynamic fashion. It is therefore key to characterize the dynamic response of the rumen microbial fermentation for identifying optimal conditions on the use of A. taxiformis as a dietary supplement for methane mitigation. Accordingly, the objective of this work was to model the effect of A. taxiformis supplementation on the rumen microbial fermentation under in vitro conditions. We adapted a published mathematical model of rumen microbial fermentation to account for A. taxiformis supplementation. We modelled the impact of A. taxiformis on the fermentation and methane production by two mechanisms, namely (i) direct inhibition of the growth rate of methanogens by bromoform and (ii) hydrogen control on sugars utilization and on the flux allocation towards volatile fatty acids production. We calibrated our model using a multi-experiment estimation approach that integrated experimental data with six macroalgae supplementation levels from a published in vitro study assessing the dose-response impact of A. taxiformis on rumen fermentation.</p>Results<p>our model captured satisfactorily the effect of A. taxiformis on the dynamic profile of rumen microbial fermentation for the six supplementation levels of A. taxiformis with an average determination coefficient of 0.88 and an average coefficient of variation of the root mean squared error of 15.2% for acetate, butyrate, propionate, ammonia and methane.</p>Conclusions<p>our results indicated the potential of our model as prediction tool for assessing the impact of additives such as seaweeds on the rumen microbial fermentation and methane production in vitro. Additional dynamic data on hydrogen and bromoform are required to validate our model structure and look for model structure improvements. Latuca sativa, cultivar Siyahoo, was selected for this experiment due to its economic value all over the world. Our studies showed that the highest yield of maize grain (8.9\u201312.0 t ha\u22121) was obtained by fertilizing with N180 and using the urease inhibitor ammonium thiosulfate (ATS). ATS significantly increased corn grain yield in all backgrounds of nitrogen fertilization. The investigated urease inhibitors and biologics had a higher and more significant (p &lt; 0.05) effect on maize grain yield when fertilized with N100 nitrogen. The increase in nitrogen fertilizer rates had an effect on maize grain yield, with the largest increase in yield being found in the increase in nitrogen rate from N100 to N140, and the increase in rate to N180 was less effective. The maximum mass of 1000 grains (323.5 g) was determined in 2019 by fertilization with N180 and use of the urease inhibitor UI URN. The urease inhibitor UI ATS was more effective when fertilized with lower rates of N100 and N140. Positive, moderate, strong and very strong, statistically significant correlations (r2 = 0.48\u20130.91) were most often found between the latter indicators and nitrogen fertilizer rates throughout the study year. The largest amount of grain (497 units) in the cob was determined in 2019, using fertilization with N140 and UI ATS, but no significant differences were found between the different fertilizer rates and the tested preparations. EnviroSCAN probes were used to determine drainage and evapotranspiration and ceramic cups to obtain soil solution samples at a soil depth of 1.4 m. The use of DMPP with urea reduced nitrate leaching. No phytotoxic effects were observed due to the DMPP. There were no differences in grain yield between treatments with and without DMPP at the same rate of N. biologics with and without methotrexate in the treatment of psoriatic arthritis: a network meta-analysis", "description": "Introduction                   <p>An important consideration in the treatment of patients with psoriatic arthritis (PsA) is whether the addition of methotrexate (MTX) to biologics has greater efficacy than biologic monotherapy with respect to efficacy outcomes in these patients.</p>                                                   Objectives                   <p>To conduct a network meta-analysis (NMA) comparing biologics by treatment class with and without MTX for treatment of adults with active PsA.</p>                                                   Methods                   <p>A systematic literature review (SLR) identified randomised, double-blinded, controlled trials, and a Bayesian NMA compared biologics with and without MTX by treatment class (tumour necrosis factor inhibitors (TNFi), interleukin-23 inhibitors (IL-23i) and IL-17i). Efficacy outcomes included American College of Rheumatology 20%, 50% and 70% (ACR20, ACR50 and ACR70) improvement response.</p>                                                   Results                   <p>The SLR initially identified 31 studies, of which 17 met feasibility criteria for the NMA by containing the \uffe2\uff80\uff98without MTX\uffe2\uff80\uff99 subgroup. For ACR20 efficacy (the most robust assessment examined), all active treatments were significantly better than placebo. No statistically significant differences were demonstrated between biologic monotherapy (for all classes examined) and biologics in combination with MTX for ACR20/50. IL-17i were comparable to IL-23i, and IL-17i were significantly better than TNFi for ACR20. Although limited by fewer trials, TNFi, IL-23i and IL-17i were not statistically different for ACR50/70.</p>                                                   Conclusions                   <p>Concomitant use of MTX and biologics did not improve ACR efficacy outcomes versus biologic monotherapy. Pain was non-radiating and worsening with movement. Patient had no constitutional symptoms. The only medication he received prior to presentation was amoxicillin/clavulanic acid as prophylaxis for a dental procedure with his symptoms starting on day 9th of therapy. Laboratory studies revealed mild leukocytosis, increased levels of serum lipase, amylase, and C-reactive protein (CRP). Abdominal computed tomography (CT) was notable for acute pancreatitis with no pseudocyst formation. Hence, patient was diagnosed with mild acute pancreatitis that was treated with aggressive intravenous (IV) hydration and pain management with bowel rest of 2\u00a0days duration and significant improvement being noticed within 72\u00a0h. On further questioning, patient recalled that several years ago he had similar abdominal pain that developed after taking amoxicillin/clavulanic acid but did not seek medical attention at that time and the pain resolved within few days while abstaining from food intake. All other causes of pancreatitis were ruled out in this patient who is non-alcoholic, non-smoker, and never had gallstones. Abdominal ultrasound and magnetic resonance cholangiopancreatography (MRCP) eliminated out the possibility of gallstones, biliary ductal dilatation, or choledocholithiasis. While urease (UI) and nitrification inhibitors (NI) are known to influence nitrogen (N) transformations, soil N availability, and utilization efficiency in ammonium sulfate urea (AS-HS), their impacts on the carbon (C) and water cycles are less understood. To address this knowledge gap and examine potential cross-effects among nitrogen, carbon, and water, we conducted an on-farm field trial with winter wheat in northeastern Germany, comparing non-fertilized, AS-HS, AS-HS+UI, and AS-HS+UI+NI treatments. Our findings indicate that inhibitors reduced N\u2082O emissions by 34.1-48.8 %. Agronomical nitrogen use efficiency (ANUE) decreased slightly with UI (1.4 %) but increased with UI+NI (4.4 %) relative to AS-HS alone. In addition to this expected effect on the N cycle, the use of UI and NI also influenced the C and water cycle by not only increasing C use efficiency (CUE) but also agronomical water use efficiency (AWUE) compared to AS-HS alone. At the same time, biomass production remained similar (UI) or even increased (UI+NI) when inhibitors were used and especially the double inhibitor treatment UI+NI seemed to effectively reduce overall global warming potential (GWP). To address this knowledge gap while reflecting realistic agronomic conditions, we conducted an on-farm strip trial in northeastern Germany, focusing on examining potential interactions among nitrogen (N), carbon (C), and water cycling. The trial compared non-fertilized plots with AS-HS, AS-HS+UI, and AS-HS+UI+NI treatments across a maize\u2013wheat\u2013barley rotation. Compared to AS-HS, the application of UI and UI+NI generally reduced cumulative N2O emissions up to 72 % and NH3 emissions up to 91 %, while it improved the N recovery efficiency (NRE) and the agronomical N use efficiency (ANUE). In addition, the use of inhibitors shifted the net ecosystem C balance (NECB) from source to sink. Although evapotranspiration (ET) increased slightly, the agronomical water use efficiency (aWUE) remained similar or even improved due to a higher biomass production. The same was observed for the net GHG emission which were similar for AS-HS and AS-HS+UI and slightly improved for AS-HS+UI+NI.
