Phytoremediation trials were conducted under real field conditions, and the thermochemical conversion of the harvested biomass was carried out at lab scale. Field trials took place in 2021\uffe2\uff80\uff932023 in Lithuania and Serbia. In Serbia, the contamination primarily involved heavy metals, whereas the Lithuanian site was predominantly contaminated with hydrocarbons from petroleum products. The harvested biomass underwent pretreatment and was then used as feedstock for conversion into high\uffe2\uff80\uff90energy carriers. The conversion products were evaluated for their potential to substitute fossil fuels. However, there has been little corresponding change in the management of nutrients, especially phosphorus (P). This study investigated the effects of tillage and crop rotations on the stratification and transformation of P in soil profiles from 3 tillage/rotation trials encompassing 3 agro-ecological zones of southern Australia. Soil samples were collected from field trials at Longerenong (Vertosol, average rainfall 420\uffe2\uff80\uff89mm), Walpeup (Calcarosol, rainfall 325\uffe2\uff80\uff89mm), and Rutherglen (Chromosol, rainfall 650\uffe2\uff80\uff89mm) in Victoria. Soil samples from various depths were sequentially analysed for organic and inorganic P fractions. Phosphorus accumulated in the surface soil (0\uffe2\uff80\uff930.1\uffe2\uff80\uff89m) across all sites and tillage practices/rotations studied but the proportion of P in different chemical fractions varied markedly among soil types and tillage practice/rotation. In the sandy Calcarosol, a greater proportion of fertiliser P was transformed into labile (resin-P) forms, whereas it tended to accumulate in non-labile pools in the finer textured Vertosol and Chromosol. The effects of tillage and crop rotation were generally confined to the topsoil with P strongly stratified in the topsoil in direct-drill and zero-tillage treatments compared with conventional tillage. These fertilizers were compared to conventional triple superphosphate (TSP) and no P fertilization (P0). We expected that reduced soil disturbance through deep loosening without soil turning would result in a higher abundance of bacteria involved in N and P turnover and an increased mycorrhizal colonization rate. Soil and rhizosphere samples were taken from two farms (Kiebitzbreite (KB) and Schmatzfelder Breite (SB)) in Central Germany during the 2018/2019 growing season when Triticum aestivum (cultivar Kredo at KB and Julius at SB) was cultivated at both sites. The abundance of microorganisms involved in P and nitrogen (N) turnover was determined using quantitative real-time PCR (qPCR) (gcd, phoD, pitA, pstS, AMF, apr, chiA, nxrA, nifH, AOA, AOB, nirS, nirK, nosZ). Three individual plants were excavated per plot for nucleic acid extraction. Soil firmly adhered to the roots was sampled as rhizosphere soil. Bulk soil samples (0 - 10 cm) were taken from the plants at a 10 cm horizontal distance. In total, 240 samples were collected (2 fields, 5 fertilization treatments, 2 soil compartments, 4 plot replicates with 3 plants each) and sieved to 2 mm. Our data show that the fertilization treatment had less impact on bacteria involved in P turnover but significantly affected the abundance of bacteria catalyzing key steps of the N cycle. Despite its simplicity and cost-effectiveness, the Kato-Katz method faces challenges, including reader subjectivity and reduced sensitivity. Real-time polymerase chain reaction (qPCR) technology offers standardized readouts and higher sensitivity, making it suitable for STH diagnosis and monitoring the treatment efficacy of emodepside within the framework of randomized controlled trials.</p>   Methodology/Principal findings <p>We evaluated the performance of Kato-Katz versus qPCR for assessing treatment efficacy in terms of cure rates, of single doses of 5, 10, 15, 20, 25 and 30\uffe2\uff80\uff89mg of emodepside compared to 400\uffe2\uff80\uff89mg albendazole. Spearman\uffe2\uff80\uff99s rank correlation coefficient examined the correlation between STH eggs per gram in stool samples and qPCR Ct values. Diagnostic sensitivity of qPCR was calculated using a Bayesian latent class modelling approach with data from Ascaris lumbricoides infections. Agreement between Kato-Katz and qPCR at baseline was 93.57% for Trichuris trichiura, and 73.49% for both hookworm and A. lumbricoides. For the latter helminth qPCR demonstrated higher sensitivity (85.00% vs. 47.70%) and slightly lower specificity (93.40% vs. 99.40%) compared to Kato-Katz. We observed a fair to moderate agreement with negative correlation between Ct values and Kato-Katz egg counts. Treatment efficacy, as assessed by qPCR, was lower for all doses of emodepside and albendazole compared to Kato-Katz. Nonetheless, emodepside demonstrated higher cure rates against T. trichiura and A. lumbricoides infections compared to albendazole.</p>   Conclusion/ Significance <p>Our study confirmed that qPCR is a sensitive diagnostic method for diagnosing STH infections compared to Kato-Katz and serves as a valuable tool for determining treatment efficacy in clinical trials. These fertilizers were compared to conventional triple superphosphate (TSP) and no P fertilization (P0). We expected that reduced soil disturbance through deep loosening without soil turning would result in a higher abundance of bacteria involved in N and P turnover and an increased mycorrhizal colonization rate. Soil and rhizosphere samples were taken from two farms (Kiebitzbreite (KB) and Schmatzfelder Breite (SB)) in Central Germany during the 2018/2019 growing season when Triticum aestivum (cultivar Kredo at KB and Julius at SB) was cultivated at both sites. The abundance of microorganisms involved in P and nitrogen (N) turnover was determined using quantitative real-time PCR (qPCR) (gcd, phoD, pitA, pstS, AMF, apr, chiA, nxrA, nifH, AOA, AOB, nirS, nirK, nosZ). Three individual plants were excavated per plot for nucleic acid extraction. Soil firmly adhered to the roots was sampled as rhizosphere soil. Bulk soil samples (0 - 10 cm) were taken from the plants at a 10 cm horizontal distance. In total, 240 samples were collected (2 fields, 5 fertilization treatments, 2 soil compartments, 4 plot replicates with 3 plants each) and sieved to 2 mm. Our data show that the fertilization treatment had less impact on bacteria involved in P turnover but significantly affected the abundance of bacteria catalyzing key steps of the N cycle.
